| Objectives:To investigate the expressions of midkine (MK) and microvessel density (MVD) in cervical cancer and to explore their relationships with clinical-pathological factors of cervical cancer.To investigate the clinical effect of neoadjuvant chemotherapy (NACT) and its influence on the expressions of MK and MVD.To discuss the feasibility and efficacy of MK and MVD's roles in predicting the effect of NACT.Methods:The expressions of MK,MVD were determined by immuno- histo chemistry in 141 cases,of which 96 were cervical cancer, 15 were high-grade squamous intraepithelial lesion (HSIL),15 were low-grade squamous intraepithelial lesion (LSIL),and 15 were chronic cervicitis.And the same method was used to detect the expressions of MK and MVD in 35 cases of cervical cancer before and after NACT.Results:1. The positive expression rates of MK in groups of chronic cervicitis, LSIL,HSIL and cervical cancer were respectively 6.67%,13.3%,40.0% and 65.6%.And the levels of MVD in these 4 groups were respectively 6.47+1.209,8.24 + 2.073,10.81 + 3.292 and 21.45+10.214, Both MK and MVD expressed higher with the development of cervical lesion (P<0.05). 2. In the group of cervical cancer the expression of MK became higher with the development of clinical stage and so did the level of MVD (P<0.05).And MK's expression was high in samples with invasion of other organs nearby (P<0.05).3. In the group of cervical cancer there was a relationship between the expression of MK and the level of MVD (P<0.05).4. Among the 35 cases treated with NACT, 4(11.43%) were complete remission (CR), 25(71.43%) partial remission (PR). The total effective rate of NACT (CR+PR) was 82.86%.5. The expressions of MK and MVD decreased dramatically after NACT.6. In cases who reacted positively to NACT, the expression of MK and the level of MVD decreased significantly after the treatment (P<0.05).While in cases who reacted poorly to NACT no such chang was observed in the expression of MK (P>0.05); but the level of MVD also decreased in those cases (P<0.05).Conclusions: Over expressions of MK and MVD were observed in cervical cancer and there was a close relationship between the two factors. NACT is an effective treatment for cervical cancer.The expressions of MK and MVD decreased after NACT. It was supposed that one of the mechanisms of NACT may be the suppression of angiogenesis, and the expression of MK may serve as an indicator for predicting the therapeutic effect of NACT. |
