Objective: By investigating the expression of PTEN,BAG-1 and GELSOLIN in the normal endometrium, atypical hyperplasia endometrium and endometrioid adenocarcinoma, this study analyzed the difference of expression among three kinds of protein and the relation between the clinicopathologic characters of endometrioid adenocarcinoma with the protein's expression in order to explore the machinery of molecular biology for endometrioid adenocarcinoma. The experience would look for the new markers for evaluating biological characters and gene therapy of endometrioid adenocarcinoma .Methods: We collected 60 cases of endometrioid adenocarcinoma tissue , 20 cases of atypical hyperplasia endometrioid tissue and 20 cases of normal endometrioid tissue . Then we examined the expression of PTEN , BAG-1 , GELSOLIN among the 100 cases of paraffin-embeded endometrioid tissue. The results was treated by Wilcoxon test , Kruskal-Wallis test and Spearman Correlation analysis .Results:1. The expression level of PTEN decreased progressively from normal and atypical endometrium tissue to endometrioid adenocarcinoma tissue. The positive immunostaining rate in the normal endometrium, atypical hyperplasia endometrium and endometrioid adenocarcinoma was 95%,45%,43.3% respectively, and the difference was significant among the three groups(P<0.01). The positive rate of PTEN was higher in normal endometrium than that in atypical hyperplasia and endometrioid adenocarcinoma(P<0.05), but there was not significance between atypical hyperplasia and endometrioid adenocarcinoma(P>0.05). There was a relationship between the expression of PTEN with the histological grade and muscle invasive depth(P<0.05), but not with age, menopause, clinical stage and lymph node metastasis(P>0.05). 2. The expression level of BAG-1 increased progressively from normal and atypical endometrium tissue to endometrioid adenocarcinoma tissue. The positive immunostaining rate in the normal endometrium, atypical hyperplasia endometrium and endometrioid adenocarcinoma was 10%,55%,78.3% respectively, and the difference was significant among the three groups(P<0.01). The positive rate was lower in normal endometrium than that in atypical hyperplasia and endometrioid adenocarcinoma(P<0.05), but there was not significance between atypical hyperplasia endometrioid tissue and endometrioid adenocarcinoma(P>0.05). There was a relationship between the expression of BAG-1 with the histological grade and lymph node metastasis (P<0.05), but not with age, menopause, clinical stage and muscle invasive depth (P>0.05).3. The expression level of GELSOLIN decreased progressively from normal and atypical endometrium tissue to endometrioid adenocarcinoma tissue. The positive immunostaining rate in the normal endometrium, atypical hyperplasia endometrium and endometrioid adenocarcinoma was 85%,45%,33.3% respectively, and the difference was significant among the three groups(P<0.01). The positive rate was higher in normal endometrium than that in atypical hyperplasia and endometrioid adenocarcinoma tissue(P<0.05), but there was not significance between atypical hyperplasia endometrioid tissue and endometrioid adenocarcinoma(P>0.05). There was a relationship between the expression of GELSOLIN with the muscle invasive depth and lymph node metastasis(P<0.05), but not with age, menopause, histological grade and clinical stage (P>0.05).4. The expression of PTEN negatively correlated with the expression of BAG-1 in endometrioid adenocarcinoma (rs=-0.512, P<0.01). There was no relation between PTEN and GELSOLIN or between BAG-1 and GELSOLIN in endometrioid adenocarcinoma (P>0.05).Conclusions:1. Expression loss of PTEN protein could accelerate the cancerization of the normal endometrium tissue .The low expression of PTEN could accelerate the histological grade and muscles invasion depth of the endometrioid adenocarcinoma .2. The decreasing expression of BAG-1 could restrain the cancerization of the normal endometrium tissue .The high expression of BAG-1 could be help to the histological grade and lymph node metastasis of the endometrioid adenocarcinoma .3. The high expression of GELSOLIN played an important restraining in the process of cancerization of the normal endometrium tissue .The low expression of GELSOLIN could accelerate the muscle invasive depth and lymph node metastasis.4. The expression of PTEN negatively correlated with the expression of BAG-1 in the endometrioid adenocarcinoma tissue. This result approved that PTEN make the function of restraining cancerization by regulating nagetively the expression of BAG-1 besides by its own action.
