HPLC is one of the most important methods to separation and analysis of the bioactive components of Chinese Medicine. However, many components have no or very weak absorption in the UV ranges such as saponins, terpenoids, lactones, glucide, and so on. Evaporative light scattering detection (ELSD) is much more suitable than UV detection (UVD) to analysis these compounds.In this thesis, analytic methods of terpenoids, saponins and lactones of Chinese Medicine were established and validated using HPLC-ELSD. The operating parameters were optimized. And these methods can be easily implemented in routine practice and used for quality control of related herbal and their preparations.This thesis mainly includes the following three parts:1) An HPLC-ELSD method for the determination of five terpenoids in . Tripterygium wilfordii has been developed and validated. The methodis simple, specific, and sensitive for the determination of triptolide, tripchlorolide, demethylzelastral, wilforlide B, and wilforlide A in different parts of Tripterygium wilfordii as well as in its preparations. The results showed that there were remarkable differences in contents of terpenoids between root, stem, leave, root bark, twig, and root without bark of Tripterygium wilfordii as well as its preparations from different companies.2) An HPLC-ELSD analytic method for determination of diosgenin in ShenMai injection was established. Diosgenin is one of the important productions that saponins of Radix Ophiopogonis in ShenMai injection transited acid hydrolyzation. Therefore, it could be applied for quality control of Radix Ophiopogonis in ShenMai injection. 3) A simple HPLC-ELSD-UVD method for the fingerprints analysis and determination of lactones and flavonoids in Ginkgo biloba has been developed and validated. The method is simple, specific and sensitive for the determination of bilobalide, ginkgolide A, ginkgolide B, and rutin in Ginkgo biloba tablets. The fingerprint chromatogram provided much more comprehensive information of Ginkgo biloba because the UV and ELSD signals contain complementary information.
