Objective1. To research the laboratory quality control of measuring acrosin activity using the gelatin lysis test in semen .2. To discuss the relationship between the perforatorium morphous and the acrosin activity in semen, to study the correlation about the different morphous in perforatorium and the acrosin activity, and the relation about the rate of fertilization in IVF.3. To research the relationship between sperm morphous and founction by transmission electron microscope(TEM).4. To choose the proper fertilization by detecting the morphous and the functional test about sperm.MethodsAccording to the standard of WHO , the samples are taken into a plastic can after Sexual abstinence 3-7days by masturbation in our special room. And then put them in 37℃to wait the liquefaction to test the routine about the sperm. We choose the samples that need to use the IVF to have a baby. The sperms in liquefaction are made the swim-up procedure. They were used to detect the sperm by the improved staining , the acrosin activity using the gelatin lysis test and the cohering experiment between the sperm and human ovule. Analyze 11 examples of sperm by transmission electron microscope.Results 1. By controling the temperature of spreading out gelatine, searching suitable density of semen and keeping gelatin plates to long time, gelatin lysis test that measures acrosin activity in semen has good stability and confidence.2. The swim-up procedure give 98.7% survival rate to spermatozoa,that provid mobile spermatozoa to check up sperm morphology and function.3. The percentage of normal sperm morphology <5% of patients have lower IVF fertility rate than normal sperm morphology≥5% patients, P<0.05,there are statistical difference.4. Sperm Acrosomal Morphology irregular or including many little vacuolus have lower fertility rate,and acrosin activity are cut down.5. There was significant statistical difference(P<0.05)in the rate of irregular acrosome between normal and disord fertilization by the test of sperm morphology.6. Analyze the relationship between sperm morphology that researched by light microscope and function by transmission electron microscope.Conclusions1. The sperm swim-up procedure can image batter sperm function and fertility rate .2. The result is reliable in using improved method of making gelatin plates and suitable density of semen .3. It is vacuole in the nucleus that was rearched by light microscope in the field of TEM. Coarctate little vacuole in the nucleus can represent spermatozoa degree of maturity4. To unite RSA, modified papanicolaou staining for spermatozoa and the acrosin activity using the gelatin analysis test can raise estimation of some off-normal sperm fertility rate.
