Objective: To study the mechanism of nitric oxide (NO) on human sperm activity and acrosome reaction function Methods: There are three parts in this trial. In the First part, high-quality sperms acquired by up-stream method were co-cultured with sodium nitroprusside (SNP) of different doses , and sperms'activity was observed by microscope at same time point. In the second part, The sperms which have capacitated, were co-cultured with SNP of different doses. The acrosome reaction by progesterone were determined by means of a flow cytometer. The FITC-PSA was used to stain the sperm acrosome which can radiate green fluorescence at 510~550nm wavelength as excited by 488nm wavelength laser. The value of FITC fluorescence was accumulated and calculated by a special computer as the stained sample passed the flow cytometer, the results were output as numeric value. In the third part , normal semen were co-cultured with SNP of different doses. To Study the change of melondialdehyde (MDA) consistency in semen. Results: 1.NO of low concentration (SNP10-9mol/L—10-8mol/L) increased the sperms'activity obviously. that of concentration (SNP10-7mol/L—10-5mol/L) had no obvious effects and that of (SNP10-4mol/L—10-3mol/L) inhibit sperms'activity marked. 2.NO of low concentration (SNP10-9mol/L —10-8mol/L) increased the sperms'acrosome reaction obviously. that of concentration (SNP 10-7mol/L—10-5mol/L) had no obvious effects and that of (SNP10-4mol/L—10-3mol/L) inhibit sperms'acrosome reaction marked.3.When SNP of concentration from 10-5 mol/L—10-3 mol/L, It will enhance MDA concentration in semen. Conclusion: Nitric oxide has been shown to play significant roles and biphasic effects in all stages of sperm motility, viability, acrosome reaction as well as lipid peroxidative. Low concentration of nitric oxide is beneficial to sperm motility, viability and acrosome reaction, and reduction of lipid peroxidative damage to sperm membranes. However, high concentration nitric oxide inhibit sperm motility, viability and acrosome reaction ,and enhances lipid peroxidative damage to sperm membranes and induces sperm toxicity. The level of biphasic effects is decided by the consistency of NO.
