Objective: To investigate the effects of AT1 receptor antagonist candesartan on acute pancreatitis(AP) of rats.Methods: 72 male Sprague Dawley rats were randomly divided into 4 groups: normal group , AP group , AP + lower dose of candesartan(2mg/ kg) group , AP + higher dose of candesartan(10mg/ kg) group , and each group contains 18 animals. AP animal models were induced by intraperitoneal injection of 20%L-arginine solution. Candesartan was given by intragastric administration. All the animals were killed in separate batches at 12h,24h and 48h(6 rats/group/batch) post-operatively. Blood was gathered by intracardiac penetration.The histopathological changes of pancreas were observed and scored(Rongione's scoring standard) , and pancreatic edema was evaluated. The levels of plasma lipase,TNF-α,IL-10 and pancreatic TNF-α,IL-10 were measured.Results :Compared with normal group , the pathologic lesions of pancreas , pancreatic edema and the levels of plasma lipase,TNF-α,IL-10and pancreatic TNF-α,IL-10 in the AP group increased significantly(P<0.01).More specifically, the pathologic lesions of pancreas and pancreatic edema increased significantly at 12 h after Arg injection, kept on increasing afterwards,and peaked at 48 h; and the serum and pancreatic TNF-a,IL-10 content increased significantly and peaked at 12 h, and then decreased but still remained elevated at a relatively constant level during pancreatitis .Compared with AP group , the pathologic lesions of pancreas and pancreatic edema decreased in the group AP +candesartan , and the levels of plasma lipase,TNF-αand pancreatic TNF-αalso reduced(P<0.05). However, there isn't significant difference between AP +low dose candesartan group and AP + high dose candesartan group(P>0.05). And the impact of candesartan on both plasma and pancreatic IL-10 is not significant in our experiment(P>0.05).Conclusion: Intragastric administration of candesartan attenuates pancreatic damage in L-arginine-induced pancreatitis on rats;...
