Object: To investigate the expression of HGF in placentas obtained from normal pregnancy and HDCP, and the effect of HGF on apoptosis of trophoblasts.Methods: 1.The microstructural changes in placentas from 30 cases of normal pregnany and 34 cases of pregnancy complicated with HDCP were detected through microscopic examination of HE stained slices.2. The expression of Fas protein were detected by immunohistochemical S-P method in placentas from 30 cases of normal pregnany and 34 cases of pregnancy complicated with HDCP.3.The expression of HGF mRNA and Fas mRNA were detected by Reverse transcription—polymerase chain reaction ( RT-PCR) in 30 cases of uncomplicated pregnancy, and 34 cases of HDCP.Results : 1.Under the microscopic examination, swelling villi, fibrinoid necrosis of villi, hyperplasia of syncytial knots and cytotrophoblast can be found. However , these microstructural changes are rarely found in the normal placenta. 2.The distribution of Fas protein in placenta of HDCP group is similar to that of normal group .Fas protein is found in villous syncytiotrophoblast and villous cytotrophoblast .Through S—P method, positive stain can be easily found on the membrane or in the plasma of trophoblast from the placenta of HDCP. Compared with normal group, the expression of Fas protein in mild preeclampsia group, severe preeclampsia group and eclampsia group were significantly higher (P<0.05), while in gestational hypertension group, there was no obvious difference(P>0.05).3.Contrast with normal group , the expression of HGF mRNA in mild preeclampsia group, severe preeclampsia group and eclampsia group were significantly lower(P<0.05), while in gestational hypertension group, there was no obvious difference(P>0.05).The expression of HGF mRNA decreased as the HDCP progressed4.Compared with normal group, the expression of Fas mRNA in mild preeclampsia group, severe preeclampsia group and eclampsia group were significantly higher (P<0.05), while in gestational hypertension group, there was no obvious difference(P>0.05).The expression of Fas mRNA increased as the HDCP progressed5.There was obvious negative correlation between expression of HGF and Fas(P<0.05)Conclusion:1.The increasing expression of Fas and the increasing apoptosis will impair the function of trophoblast. Furthermore, that will defect the supplication of blood and oxygen to placenta, and result in HDCP.2.Compared with normal group, the expression of HGF mRNA decreased as the HDCP progress, which indicates that HGF may play an important role in HDCP.3.HGF secreted by mesenchyme inhibits the apoptosis mediated by Fas , and reduction of HGF in HDCP may be responsible for apoptosis.
