| Objective: Transforming growth factor-β1 (TGFβ1) is one of the most multi-functional growth factors,it gains more attention in recent years.The most important and characterristic function is stimulate proliferation of trophocytes.Obviously correlation was found between TGFβ1 concentration and level of blood pressure (P<0.05) in patients with hypertensive disorders in pregnancy.The expression of TGFβ1 may have a key role in embryo growing.Placental growth factor(PLGF),a member of vascular endothelial growth factor family,is mainly expressed in normal placenta and secreted prominently in trophoblasts.It can stimulate proliferation of trophocytes and mediate angiogenesis.Its receptor flt-1 is located on the surface of trophocytes and endothelial cells.Placental growth factor regulates the functions of trophocytes and vascular eudotheli cells,so it plays key role in placenta formation and development of normal pregnancy.Pregnancy with hypertension is called hypertensive disorders in pregnancy(HDCP).It is one of the complications that are often encountered in obstetrics. Hypertensive disorders in pregnancy is associated with abnormally shallow cytotrophoblast invasion.It's vascular remodeling in the spiral arteties is confined to the decidual portion.Managment of hypertensive disorders in pregnancy has changed dramatically over years and a conservative approach now predominates.In order to allow a conservative treatment,accurate cause and mechanism are important.To investigate the expression of transforming growth factor-β1and placentaL growth factor in placentae of pregnancy-induced hypertensive disorders in pregnancy patients and their significance.Methods: This was aprospective,case-control study ebaluation TGFβ1 concentrations and PLGF expression.Normal group was consisted of 30 normal pregnancies.HDCP group was consisted of 58 women.A total 1.0×1.0×1.0cm of placenta tissue was withdrown as soon as it was delivered.The placenta tissues were routinely fixed in 10percent formalin and embedded in paraffin.Four-μm-thick tissue sections of all samples were stained by immunohistochemical analysis.Positive expression evaluation was used to asses the immunostaining intensity of the proliferation of trophocytes.Analysis of two-sample-test and spearman rank correlation were used to analyze with spass 12.0 software,P<0.05 was considered statistically signigicant.Results: 1 To investigate the localization of TGFβ1 and PLGF in normal pregnancy: TGFβ1 and PLGF was found in syncytiotrophoblast and cytotrophoblast(Fig1,2).2 To investigate the localization of TGFβ1 and PLGF in hypertensive disorders in pregnancy patients:Same to the 1th.But the colour of the stained cells was stronger than the contrlo group(Fig3-6).3 To investigate the evaluation of TGFβ1 and PLGF:In the normal pregnancy group,the expressions of TGFβ1 and PLGF were: 33.33﹪,46.67﹪, the expressions in the mild HDCP were:51.52﹪,39.40﹪, the expressions in the serve HDCP were:82.86﹪,14.29﹪.The expression of TGFβ1 in placental trophoblasts and decidual cells was significantly increased in the HDCP group compared with the normal group(P<0.05),and the expression in the serve HDCP group was higher than that in the mild HDCP group(P<0.05);The expression of PLGF was significantly decreased in the HDCP group (P<0.05)(Table1) ,and the expression in the serve HDCP group was lower than that in the mild HDCP group(P<0.05).Negative relationship was shown between the expression of TGFβ1 and PLGF.4 In patients with severe preeclampsia,TGFβ1 expression showed positive correlation with PLGF expression (r=-0.350 p<0.05).Conclusion: Based on the data mentioned above,we can come to the following conclusion: TGFβ1 and PLGF may play the important role in HDCP and maintenance of pregnancy.The expression of TGFβ1 in placenta of normal pregnancy group is weaker than the HDCP group,and the expression of PLGF is stronger.So we can measure the expression of TGFβ1 and PLGF in placental trophoblasts and decidual cells to diagnose hypertensive disorders in pregnancy.At the same time,associating of them can increase the early diagnosis significance. |
