Objective To detect the mutation in two exons of BRCA1 gene in sporadic ovarian carcinoma, and to study the relationship between BRCA1 mutation and bteast carcinoma. Methods A total of 36 patients with ovarian carcinoma without blood relationship were randomly selected. Polymerase chain reaction(PCR) Single-strand conformational polymorphism(SSCP) was used to test the mutation of exons 2,20 of BRCA1. Results No mutations were found in exons 2 and 20. Conclusion 185 delAG and 5382 insC which are high mutation sites in Ashkenazi Jewish women were not high in China;the result indicate that gene sequence of what we have studied doesn't account much for occurrence of the sporadic ovarian carcinoma in the population of china.It is important to find new mutations in the sporadic patients of ovarian carcinoma in China. Objective To explore the clinical significance of c–erbB-2 oncogene amplification in ovarian carcinoma. Methods c-erbB-2 oncogene amplification was examined by using differential polymerase chain reation in 36 cases of ovarian carcinoma. Results (1) c-erbB-2 amplification was found in 58.3%(21/36)of ovarian carcinoma. (2) The positive expression rate of c-erbB-2 in patients with≥5cm tumor size and late stage were 67.9%,73.9%,were higher than that in﹤5cm tumor size (25.0%)and early stage(30.8%).(3)c-erbB-2 oncogene amplification was not correlated to age and histologic grade. Conclusion The results suggest that c-erbB-2 amplification play a role in the process of ovarian carcinoma and may be one of those significant prognostic factors for therapy,predicting prognosis for ovarian carcinoma.
