Objective To observe the change of tumor blood supply and expression of VEGF , MVD after ligating the hepatic artery to supply the tumor in rabbit hepatic VX2 carcinoma, initial investigate the blood source of residual focus and its change of molecular biology, guiding clinic to further treat primary hepatic carcinoma after multiple TACE. Materials and Methods 40 New Zealand White rabbits ,body weight 2.5~3.0 kg ,without thinking their sexualities,divided randomly into Group A and Group B. Hepatic artery DSA were performed on 3 weeks after VX2 tumor implantation in livers . Group A were performed the celiac trunk and portal vein DSA at 24 h,72 h and day 14 after ligating the hepatic artery to supply the tumor. Group B without hepatic artery ligation do it like group A at the same time. To observe if the tumor has anther artery blood supply and collateral circulation ,then to measure the diameter of main portal vein and to number the fourth branch of portal veins which were pressed by tumor body . Each six rabbits were sacrificed at 24 h,72 h and day 14 after performed the celiac trunk and portal vein DSA and tumor tissues were excised. Expression of vascular endothelial growth factor (VEGF) and micro vessel density (MVD) protein of tumors were examined using immunohistochemistry. Results The making rabbit VX2 tumor model success rate was 92.5%.All tumors were supplied by hepatic artery. There were significant differences about diameter of main portal vein and the number of fourth branch of portal vein between the 24 hours and day 14 after hepatic artery ligation , P<0.01.The perimeter of tumors were stained on day 14 after HAL treatment through portal vein DSA. The differences were significant between the 24 hours and 72 hours about VEGF, P<0.01, expression of VEGF in the central region of tumor descended obviously on day 14. MVD heightened obviously on day 3 after HAL, and descended obviously on day 14 in the central region of tumor, there were significant differences with day 1, P<0.01. There were significant correlation between VEGF and MVD. Conclusions The method of making rabbit hepatic VX2 carcinoma model is simple, and the success rate is very higher, and the New Zealand White rabbits were very fitted to operate interventional techniques. The central region of tumor was supplied completely by hepatic artery, and the perimeter of tumor was supplied by portal vein, when the tumor was lost blood supply by artery.
