Objective To compare the protective effect of Shaoqiduogan using the different extraction methods on immunological liver injury in mice.Methods The model of immunological liver injury in mice was produced by injection of Bacillus Calmette Guerin( BCG) and lipopolysaccharide ( LPS ) from tail vein. The levels of alanine aminotransferase(ALT), aspartate aminotransferase(AST), malond-iadehyde(MDA)content, superoxide dismutase(SOD) and glutathione peroxidase (GSH-Px) activities in liver homogenate were assayed by spectrophotometry, tumor necrosis factor-α(TNF-α) activity was determined by radioimmunoassay. The level of interleukin-1(IL-1)was assayed by MTT dye reduction. Hepatic tissue sections were stained with hematoxylin and eosin and examined under a light microscope.Results (1) SQDG (60,120,240mg·kg-1) and TGP+ASTs (52.5+7.5,105 + 15,210 +30mg·kg-1) were able to significantly decrease liver index, spleen index and decrease serum transaminase (ALT,AST)levels of immunological liver injury's mice induced by BCG plus LPS, attenuate the area and extent of necrosis and reduce the infiltration of inflammatory cells.Furthermore, SQDG (120,240mg·kg-1) and TGP+ASTs (105 + 15,210+30 mg·kg-1) also decreased MDA content and improved the reduced SOD and GSH-Px levels in liver homogenate. Meanwhile, SQDG (60,120,240mg·kg-1) and TGP+ASTs( 52.5+7.5,105 + 15,210+30mg·kg-1 inhibited the production of TNF-αand IL-1 by peritoneal macrophages. (2) Comparing with TGP+ASTs (105 +15mg·kg-1), SQDG (120mg·kg-1) significantly further changed the serum transaminase levels, the production of MDA and TNF-αwhile had no difference effects on other indices.Conclusion SQDG and TGP+ASTs possess protective action on immunological liver injury in mice. SQDG (120mg·kg-1) and TGP+ASTs (105+ 15mg·kg-1) have pharmic difference on the model of immunological liver injury in mice.
