Shaoqiduogan (SQDG) is an effective compound extracted and purified by modern techniques from Radix Paeonia Pall and Radix Astragali with the optimum dose proportion. It has been proved that SQDG played a protective role in immunological and chemical liver injury of rats, and also had modulatory effect on some cytokines. To investigate the effects of SQDG on immune system, this study researched the effects and mechanisms of SQDG on cellular immunity of mice.Objective To investigate the effects of Shaoqiduogan (SQDG) on cellular immunity of mice with delayed-type hypersensitivity (DTH), and the comparison of effects of SQDG, total glucosides of paeony (TGP) plus astragalosides (ASTs) and TGP on cellular immunity.Methods DTH model induced by 2,4-dinitrochlorobenzene (DNCB) and cyclophosphamide (Cy) suppressed and potentiated DTH model were used . SQDG,TGP,ASTs and TGP plus ASTs were given intragastrically ( ig ) once daily for 7 consecutive days. Ear swelling, thymus and spleen index of mice were investigated. Concanavalin A (ConA)-induced T cells proliferation was measured by MTT assay. The activity of IL-2 in culture supernatants of ConA-induced T cells proliferation was measured by testing its ability to support the proliferation of ConA stimulated thymocytes which was detected by MTT assay.Results (1) In DTH mice and Cy-potentiated DTH mice, SQDG (60,120 mg·kg-1) reduced ear swelling, thymus and spleen index, ConA-induced T cells proliferation and the activity of IL-2 in culture supernatants of ConA-induced T cells proliferation. SQDG (60,120 mg·kg-1) improved ear swelling, thymus index, ConA-induced T cells proliferation and the activity of IL-2 in Cy-suppressed DTH mice.(2) In DTH mice, comparing with TGP+ASTs (52.5+7.5) mg·kg-1, SQDG (60 mg·kg-1) reduced thymus and spleen index, ConA-induced T cells proliferation and the activity of IL-2; SQDG (120 mg·kg-1) was superior to TGP (140 mg·kg-1 ) in decreasing the activity of IL-2. In Cy-suppressed DTH mice, comparing with TGP+ASTs (26.25+3.75, 52.5+7.5, 105+15 mg·kg-1), SQDG (30, 60, 120 mg·kg-1) improved the activity of IL-2. In Cy-potentiated DTH mice, comparing with TGP+ASTs (26.25+3.75, 52.5+7.5 mg·kg-1), SQDG (30, 60 mg·kg-1) decreased ear swelling. SQDG (30, 60, 120 mg·kg-1) were superior to TGP+ASTs (26.25+3.75, 52.5+7.5, 105+15 mg·kg-1) and TGP (70, 140 mg·kg-1) in inhibiting T cells proliferation and the activity of IL-2. Conclusion SQDG possesses a dual modulatory effect on cellular immunity in mice. There were pharmic differences between SQDG and TGP+ASTs, TGP in modulating cellular immunity of mice.
