ObjectiveThe concept of kidney fibrosis is under the effect of various causative agent induce glomerular sclerosis and nephric tubule interstitial fibrosis. At present,there is no effective intervention study to delay the progress of kidney fibrosis. The objective of this experiment is through observe the influence of calcium dobesilate to the CRF model rat 's nephridial morphologic and the express of TGF-β1 and VEGF in nephridial tissue, to approach the mechanism of action of calcium dobesilate improve the CRF model rat's kidney microcirculation and delay kidney fibrosis.MethodsTwenty eight SD rats ran divided into therr groups: eight rats in control group, ten rats in calcium dobesilate group, ten rats in not treat group. In four weeks, make the model of CRF rats through adopt operation and twice intravenous injection A driamgcin to the rats in the calcium dobesilate group and the not treat group. During the fifths and the sixth week to give calcium dobesilate group rats calcium dobesilate (An duo ming ) 100mg/KG·d lavage tow weeks. Control group and not treat group mitte talec doses normal saline.The sixth week of the experiment to put all rats to death and to weigh, get abdominal caval vein blood, determin serum creatinine ,blood urea nitrogen, removal kidney and to weigh, fixed, observe with light microscope, immunity tissue chemistry determin to TGF-β1 and VEGF.ResultsSerum creatinine and blood urea nitrogen of calcium dobesilate group rats were obviously hyper than control group rats. The difference between not treat group and control group have statistical significance (P<0.01) .serum creatinine and blood urea nitrogen of calcium dobesilate group rats were obviously lower than not treat group rats (P<0.01). HE,PAS,PASM,Masson dyeing of not treat group rats kidney tissue can see most Mesangial cell height hyperplasia, extracellular matrix mult, saccuius proprius adhesion, part glomerular capillary completely blunting innings focus S-T segment S-T hardening change, part renal tubular epithelial cells vacuolar degeneration, intralumens have protein cast.Renal interstitium muiti-focus-form cell infiltrate, part nephric tubule-interstitial fibrosis. Acinus renis fibrosis of calcium dobesilate group was obviously abatement than not treat group and intercapillary cells and mesenterterium base material obviously decrease, spouse to see sacculus proprius adhesion and S-T segment S-T fibrosis, a few kidney lumens have protein cast, kidney interstitial fibrosis not obviously. Immunity tissue TGF-β1 dyeing of control group only gleam express inside a few renal tubular epithelial cell, not treat group obviously hyper than control group, TGF-β1 dyeing express in calcium dobesilate group obviously decrease than not treat group (P<0.01) . VEGF dyening express in not treat group was obviously lower than control group. VEGF dyening express in calcium dobesilate group was obviously accrescence than not treat group (P<0.01) .ConclusionCalcium dobesilate can abaissement the composition of nephridial tissue region TGF-β1 Calcium dobesilate can amelioration the level of nephridial tissue fibrosis of CRF rats. Calcium dobesilate can accrescence nephridial tissue the express of VEGF and improve kidney partly microcirculation . Calcium dobesilate obviously to lower CRF rats Scr and BUN, amelioration renal function.
