Objective:To study the scavenging effect of YLS polysaacharide and total saponins on superoxide anion free radical(O2·-)and hydroxy free radical (·OH).Methods:Spectrophotometry(wave length 322 nm and 510 nm)was used to observe the effects of YLS polysaccharide and total saponins on O2·- and·OH.Results:The level of O2·-was reduced by 38.5%,49.1%and 58.9% in the presence of YLS polysaccharide at 0.25,0.5,and 1.0 mg·ml-1in vitro,the generation of O2·-was inhibited by 29.7%,38.1%and 55.3%,respectively, and the level of·OH was reduced by 24.2%,35.6%,and 56.9%,respectively. The level of O2·-was reduced by 7.8%,23.8%and 44.6%in the presence of YLS total saponins at 0.15,0.3,and 0.6 mg·ml-1in vitro,and the generation of O2·-was inhibited by 17.1%,33.4%and 41.3%respectively,but the level of·OH was reduced by -11.9%,-18.4%,and -32.2%, respectively.Conclusion:YLS polysaccharide and total saponins can greatly scavenge O2·-,and inhibit the production of O2·-.YLS polysaccharide can also scavenge·OH,while YLS total saponins can increase the generation of·OH. Objective:To observe the protective effect of Yulangsan(YLS)polysaccharide on focal cerebral ischemia/reperfusion(I/R)injury in rats.Methods:Wistar male rats were randomly divided into I/R group,sham-operated group,high-, medium- and low- dose of YLS polysaccharide groups(0.6,0.3 and 0.15 g·kg·d-1respectively),and nimodipine(Nim)group.The animals were intragastrically administered with drugs for 7 days.The operation was performed to induce the middle cerebral artery occlusion(MCAO)model in rats with the method of intraluminal thread at 60 min after the end administration of drug.After ischemia for 2 h and reperfusion for 24 h in each model rats,the influences of YLS polysaccharide on the neurological function score,the brain water content,the infarct volume,the activities of SOD,NOS and ATPase,the contents of MDA and NO,the expressions of Bcl-2 and Bax,and the concentration of TNF-a and IL-βin brain tissue were investigated;the morphological changes of rat cerebral cortical neurons(HE staining)were observed under optical microscope.Results:Compared with I/R group,YLS polysaccharide could obviously reduce the neurological function score,the brain water content,the infract volume,MDA and NO contents,NOS activity,the expressions of bax,and the concentration of TNF-a,IL-β,and increase SOD and ATPase activity.The express of bcl-2 in the brain tissue(P<0.01 or P<0.05)and the neuronal edema could be lessened.Conclusion:YLS polysaccharide has protective effect on cerebral ischemia/reperfusion injury,which mechanism may be related to attenuating free radical;increasing ATPase activity,up-regulating Bcl-2 and down-regulating Bax,increasing the Bcl-2/bax ratio,inhibiting the expression and release of the inflammatory mediators.
