Objective: To study the effect of ELHE on inflammatory response after focal cerebral ischemia by investigating the changes of TNF-α,IL-1β,MCP-1 and NF-κB in brain tissue . Methods: Brain ischemia model was induced utilizing the local injection of self-thrombus into the middle cerebral artery with sterile technique. ELHE was used as the intervension drug.At different time points, NF-κB was detected by immunohistochemistry, TNF-α,IL-1βand MCP-1 levels by ELISA,and the level of inflammation in ischemic brain tissue by HE. Neurological severity scores was evaluated by Berderson's method. Results: 1. By HE staining,very few leukocytes was observed in sham group (SG); In model group (MG), the number of leukocytes increased markedly at 1st day, reached the maximum at 2nd day,then decreased. In Shexuetong-treated group(ETG), there was a significant decrease at each time point,especially at 3rd days. 2. In MG, the NF-κB positive cells in the ischemic brain tissue(IBT)reached the peak at 1st day,began to decrease slightly at 2nd day and 3rd day, decreased significantly at 5th day, there was a remarkable difference compared with each time point of SG (P<0.05); In ETG, the rule of the positive expression was similar to the MG,ELHE significantly reduced the positive cells of all the time points (compared with MG, P<0.05). 3. In MG, the content of TNF-αin the IBT increased markedly at 1st day, reached the peak at 2nd day,decreased slightly at 3rd and 5th day, which had a remarkable difference compared with each time point of the SG(P<0.05); In ETG, ELHE reduced the content of TNF-αin the IBT, especially at 3rd day(compared with MG, P<0.05). 4. In MG ,the content of IL-1βin the IBT increased markedly at 1st day, the peak at 2nd day, decreased slightly at 3rd day, markedly at 5th day(compared with SG, P<0.05). In ATG, ELHE reduced the content of IL-1βof each time point,especially at 2nd and 3rd day(compared with MG, P<0.05). 5. In MG ,the content of MCP-1 in the IBT increased markedly at 1st day, the peak at 2nd day, still remained high level at 3rd day,decreased markedly at 5th day (compared with SG, P<0.05); In ATG, ELHE reduced the content of MCP-1 of each time point(compared with MG, P<0.05).Conclusion: ELHE can alleviate the leukocyte infiltration,inhibit the expression of NF-κB, reduce the protein level of TNF-α,IL-1βand MCP-1 following MCAO.So ELHE play a protective role in inflammatory response after brain ischemia, which probably relates to inhibiting the protein expression of NF-κB,TNF-α,IL-1βand MCP-1 following MCAO. Objective: To study whether ELHE protects ischemic brain tissue from the injury induced by oxygen stress, we observed the changes of MDA ,SOD and NOS in ischemic brain tissue by the intervension of ELHE. Methods: Brain ischemia model was induced utilizing the local injection of self-thrombus into the middle cerebral artery with sterile technique. ELHE was used as the intervension drug. At different time points, the level of MDA was detected by TBA method, the activities of SOD and NOS by biochemical method. Results: 1. In MG, the content of MDA in the IBT increased greatly at 1st day, reached the peak at 5th day,which has a remarkable difference at each time point compared with SG (P<0.05); The content of MDA in ETGremarkabaly decreased at each time point compared with MG(exclude 1st d, P<0.05). 2. In MG, the activity of SOD in the IBT was the lowest at 1st day, then increased lightly at 2nd and 3rd day, reached the peak at 5th day,which has a significant difference with SG(P<0.05). Compared with MG, the activity of SOD in ETG remarkably increased at each time point(P<0.05). 3. In MG, the activities of NOS and iNOS in the IBT increased greatly at 1st day , reached the peak at 5th day,then dicreased gradually, compared with SG , the difference between each time point was remarkable (P<0.05); Compared with MG, the activities of NOS and iNOS in ETG remarkably decreased at each time point(P<0.05).
