Objcctive:To Study the expresion level and distribution of human beta-defensin-3(To call for thorth:HBD-3) in chronic tonsillitis tissue and normal amygdala tissue. To investigate the effect of HBD-3 in the infection aspect of chronic tonsillitis and provide the theory evidence for the clinic biotherapy of chronic tonsillitis and the preparation of biologic agent.Methods: with twenty six postop. stissue pecimens of chronic tonsillitis patients and ten coloretured normal amygdala tissue pecimens of obstructive sleep apnea hyponea syndrome(normal control) patients, by the hematoxylin-eosin colouring(HE) and Streptavidin -Peroxidasemethod (S-Pmethod) of immunohistochemistry with monoclonal antibodyes against HBD-3, To observe the pathological change of the tonsillitis and research the expression position of HBD-3 in the tonsillitis tissue and investigate quantitative change of HBD-3 in amygdala tissue of two groups patients. The data obtained by the computer image processing system was analyzed withstatistics means.Rsults: Observe HE slice under the light microscope, The distinctive pathologicalchange is fibrous connective tissues of proper layer hyperplasia in the normal control group. But in the experimental group, The distinctive pathological change is lymph foilicie hyperplasia, germinal center enlargement, inflammatory cell infiltrate in the epithelial lamina and the small amounts proteinum exudation. To observe the immunohistochemistry slice under the light microscope: HBD-3 expression in cell membrane and endochylema of the nonkeratinized stratifired squamous epithelial both in the experimental group and normal control. The HBD-3 expressive position is identical in both groups. The semi-quantitive analyses have been shown that the HBD-3 expression are positive or strongly positive in 26 specimens of the experimental group; However In 10 specimens of the normal control groups, 7 specimens are weakly positive, 2 specimens are negative ,1 positive. The statistical analysis shown HBD-3 expression is significant deviation in both groups(P<0.01). Immunohistochemistorical results of HBD-3 in all specimens sections: the general grey averages are 45. 9450±16. 97154 and 87. 6100±14. 07553 in the normal control group and the experimental group respectively (p<0.01).Conclusion: HBD-3 is inducible expression in chronic tonsillitis tissue. The results suggest that the epithelial cell of the palatine tonsil excrete beta-delensin-3 may play an important role in innate defense against microorganisms.
