| Objective:To explore the Association between two syndromes(phlegm and blood stasis syndrome and predominant liver yang syndrome)defined by traditional Chinese medicine in cerebral infarction(CI)and angiotensinogen(AGT)gene N235T polymorphism.Nethod:All subjects comprised 104 CI patients and 50 health controls aged 50-80 years,the patients are divided into 2 groups at random,group A is comprised 56 cases had been diagnosed as phlegm and blood stasis syndrome defined by traditional Chinese medicine,and group B is comprised 48 cases had been diagnosed as predominant liver yang syndrome defined by traditional Chinese medicine.Otherwise recorded the informations of risk factors of all the cases.M235T polymorphism of AGT gene from peripheral blood leukocytes was examined in 104 cases with CI controls and 50 cases with health controls by polymense chain reaction(PCR)amplification and restriction fragment length polymorphism(RFLP).And recorded the results.Result:1,Angiotensinogen(AGT)gene amplified by PCR produce a DNA fragment Of 163bp.The product of PCR was digested by restriction enzymes Tth111 -â… shown that there was a DNA fragment of 163bp,140bp or a DNA fragment of both 163bp and 140bp.2,Comparison of risk factors of cerebral infarction between two groups on,there was only a statistical significance on the blood pressure (P<0.05).3,Compare with 104 cases with CI controls and 50 cases with health controls by PCR-RFLP,the frequencies of AGT-TT genotype and T allele in CI group are both higher than that in health group(X~2=5.77 P<0.05;X ~2=9.75 P<0.01);the frequency of AGT-TT genotype in group B was higher than health group(X~2 =5.95 P<0.05);but there was no statistical difference in the frequencies of AGT genotype and allele between group A and group B(X~2=1.56,P>0.05;X~2=1.14,P>0.05).conclusion:There was a relationship between CI and AGT-TT genotype and T allele(X~2=5.77 P<0.05;X~2=9.75 P<0.01);the predominant liver yang syndrome defined by traditional Chinese medicine maybe contact with AGT-TT genotype(X~2=5.95 P<0.05);there was no obvious statistical difference between the two groups in frequencies distributes of AGT gene genotype and allele(X~2=1.56,P>0.05;X~2=1.14,P>0.05),and the conclusion was similar with the previous studies.Otherwise,blood pressure as risk factor among group A compared with those among group B, There was statistical difference between the two groups(P<0.05),it means that there might be diffence between predominant liver yang syndrome and others in the performance of blood pressure. |
