| Objective: To analyze the expression of caudal-related homeodomain transcription factor 2(CDX2) and mucin 2(MUC2) in the chronic inflammation of cardia, short segment Barret′s esophagus (SSBE) and adenocarcinoma of cardia, and study the biological effect of them.Methods: The study consists of 35 biopsies of reflux esophagitis,30 biopsies of SSBE, 26 biopsies of chronic inflammation of cardia (no intestinal metaplasia 3, low intestinal metaplasia 10, moderate intestinal metaplasia 8, high intestinal metaplasia 5),5 biopsies of normal esophageal and 5 biopsies of normal cardia as normal control,5 biopsies of normal colon as positive control. Using immunohistochemistry, the expression of CDX2 and MUC2 were detected in every group. Statistical analysis was performed using SPSS 13.0 software.Results: In reflux esophagus, the positive rate of CDX2 and MUC2 expression were 22.9%(8/35)and17.1%(6/35), in SSBE were 76.7%(23/30) and 66.7%(20/30), in chronic inflammation of cardia were 73.1%(19/26) and 69.2%(18/26),in adenocarcinoma of cardia were 52.6%(10/19),47.4%(9/19).The expression of reflux esophagus were lower than those of other groups (P<0.05).There was no difference between SSBE and chronic inflammation of cardia(P>0.05).The expression of adenocarcinoma of cardia groups was lower than that of SSBE and chronic inflammation of cardia(P<0.05).There was no expression of CDX2 and MUC2 in chronic inflammation of cardia with no intestinal metaplasia; the expression in low intestinal metaplasia group,moderate intestinal metaplasia group and high intestinal metaplasia group were:(31.05±11.13,19.90±15.19),(30.23±9.34,29.94±11.63),(77.13±12.07,73.86±13.15).No difference was found between low intestinal metaplasia group and moderate intestinal metaplasia group(P>0.05)the positive expression intensity of high intestinal metaplasia group was significantly higher than those of low intestinal metaplasia and the moderate intestinal metaplasia groups(P<0.01).Conclusion: CDX2 was one of important gene that induced normal cell to intestinal metaplasia.CDX2 could modulat the expression of MUC2. The chronic inflammation of cardia with high positive rate expression of CDX2 and MUC2 might be a significant precancerous state.CDX2 might be involved in the development of adenocarcinoma of cardia. |
