AIMS:The aims of present study are to establish insulin resistance model in vivo and vitro and observe the effects of betaphrine on insulin resistance. METHODS:(1) In vitro study, HepG2 cells exposed to 5×10-7M insulin for 16 hours, glucose consumption was determined which was used to estimate insulin sensitivity of HepG2 cells. After establishment the insulin resistance HepG2 cells model,the HepG2 celsl were incubated with betaphrine(1×10 -6~10 -4M) for 24h,glucose consumption was determined,Western blotting was used to assess the levels of GLUT-4.(2) In vivo study, DM(1mg .kg-1 )was injected every other day for 3 weeks to induce rats insulin resistance. After insulin resistance model of rats are reproduced,experimental group was treated with betaphrine.the experiment lasted 3 weeks . Results were evaluated by measurement of serum lucose,serum insulin, serum triglyceride ,hepitac glycogen and GLUT-4.Insular structure was to observe by HE dyeing.RESULTS:(1) In vitro study, the glucose consumption of HepG2 cells significantly decrease by coincubated with 5×10-7M insulin for 16 hours compared with control(P<0.01)。Compared with model group, betaphrine significantly increase the glucose consumption of insulin resistance HepG2 cells in a manner of concentration-dependance(P<0.05), increase the ecpressed of GLUT-4(P<0.01).(2) In vivo study, the serum glucose of rats significantly increase by injected with DM(1mg .kg-1 ) for 3 weeks compared with control(P<0.01).Compared with model group, betaphrine decrease the levels of serum glucose , serum insulin, serum trilyceride and hepitac glycogen in a manner of concentration-dependance(P<0.05), increase the ecpressed of GLUT-4(P<0.05).CONCLUSIONS:(1) HepG2 cells exposed to 5×10-7M insulin for 16 hours were able to induce a state of insulin resistance. Betaphrine significantly increase the glucose consumption of insulin resistance HepG2 cells,improved the insulin resistance in HepG2 cells.(2) Rats injected with DM(1mg .kg-1 ) for 3 weeks were able to induce a state of insulin resistance. Betaphrine is a anti-diabetic effects maybe associated with inceease the expressed of GLUT-4.
