The Research Of The Chemical Constituents From The Bioactivity Part And The Quality Specification Of Chuanmingshen

Chuanmingshen violaceum Sheh et Shan, a traditional Chinese herbal medicine, belongs to Umbelliferae family. It is only distributed in China as Sichuan authentic herbal medicine. Chuanmingshen has the traditional effects of moistening the lung and resolving the phlegm, militate the stomach, promote secretion, detoxification ,etc. In clinic, it is frequently applied to treat cough caused by lung-heat, consumption of yin caused by febrile disease and so on. It is widely used in treatment for nourishing Yin both as food and drug.As the Sichuan authentic material, Chuanmingshen serves both as medicine and food, which has a large market. But until now seldom researches on its chemical components and bioactivities have been done, so there is no any systematic and. standard specification to control the quality. The only quality control standard of this material is the simple appearance description written in the (87 section), and Chuanmingshen hasn't been adopted by the < Chinese Pharmacopoeia> 2005ed Vol.1 until now.For the aim of exploring the precious Sichuan material, promoting the development of the local economic, clarifying the bioactivity mechanics and finding the quality control indicator, we initiated a study of the pharmacological effects and the chemical components of Chuanmingshen. Firstly, by means of 4 pharmacological models(a. ammonia liquor induced coughing experiment, b. tracheal phenol red experiment, c. carbon granular clearance experiment, and d. serum hemolysin experiment, in mice.), we found that the petroleum ether extract is the bioactivity part of Chuanmingshen for antitussive, expectorant effects, and the humoral immunity influences which was ensured by another vitro experiment: T lymphocyte proliferate experiment by the MTT assay, done after. Based on this conclusion, we sequently studied on the chemical components from the petroleum ether extract of Chuanmingshen, isolating 8 compounds from this extract, Hexadecanoic Acid (A), 2,6-Dimethoxybenzoic Acid (B), Succinic acid (C), combination of Sigmasterol and Sitosterol(1: 1) (D), Chrysophanol(E), 8-Hydroxy-5-methoxypsoralen(F),4-Hydroxy-3-methoxybenzoic acid(G), and stigmasteryl 3-O-β-D-glucopyranoside(H), B,C,D,E,G were isolated from Chuanmingshen for the first time.Based on these results, we studied on the standardized quality specification of Chuanmingshen basically. We took the psoralen, 8-Hydroxy-5-methoxypsoralen, and Chrysophanol as the reference compound to set up the TLC identification method and setted up the content assay method of the total quantity of coumarin of Chuanmingshen by means of the UV-VIS spectrometer.These results of our study provided a research foundation for finally revealing the bioactivity substance and founding the standard quality specification of Chuanmingshen, thus will broaden the clinical application and the new medicine R&D of Chuanmingshen.
