| Medicinal Chuanminshen is the plant of umbelliferae Chuanminshen violaceum Sheh et Shen genus and Si Chuan famous-region frug,it can nourish yin and tonify the lung,fortify the spleen,it can treat the syndrome of cough with lung heat and consumption of YIN caused by febrile disease.Mainly contain polysaccharide and coumarins components,for the traditional Chinese medicine and medicine dual-use plants.With the increase in market demand,high quality resources and improve the quality of the system more and more attention.Based on the existing research,this paper chooses Sichuan Chuanming as the research object,through the investigation of the biological traits and biomass of Chuanminshen resources,screening the high-quality resources;To carry out experimental research and comparison analysis for the quality characteristics of chuanminshen,discussed the difference between quality characteristics and active ingredients in different producing areas,and discussed the the distribution of characteristic components in the different parts and structures.Ues the single factor experiment and orthogonal test to optimized for the extraction of polysaccharises and coumarins from chuanmingshen;Determine the best origin processing methods and established a multi-dimensinal characteristic map of chuanmingshen;And on this basis,screening high quality chuanmingshen resources,the establishment of quality standards.The main findings are as follows:1.The biological characters and biologic yield of Chuanming ginseng were determined and analyzed statistically.The differences in the biological traits and yield of each resource type were determined.Then analysised the relationship between biological characteristics and yield of aboveground and underground parts of different resource types.Established related regression equation,build a bridge between yield and trait parameters.Preliminary studies show that:The root circumference is wide leaf type(8.17cm)>fine leaf type(7.10cm)>lobular type(6.83cm);The average root weight is lobular type(112.62g)>fine leaf type(92.35g)>wide leaf type(72.27g);Plant roots on the premium and level of sensory quantity grading composition(1-3)root rate is lobular type(14.71%;79.42%)>fine leaf type(10.81%;67.57%)>wide leaf type(6.38%;65.95%).There were significant correlations between the aboveground biological characters and biomass of chuanminshen(P<0.05*),Lobular Chuanming is a high quality and high quality germplasm resources.2.Experimental Study on Extraction Technology of Polysaccharides and Coumarins for chuanminshen:The optimum extraction process of polysaccharides and coumarin from Chuanming was determined by single factor and orthogonal experiment.Polysaccharides of optimum extraction conditions were as follows:water as solvent,ratio of material to liquid 1:40,extraction temperature 100 ?,extraction time 2.5h,alcohol concentration 80%,extraction times 2 times;Coumarin extraction of the best process:methanol as solvent,ultrasonic 30min,temperature 50?,ultrasonic power 80W,material ratio 1:20.3.The experimental results show that:(1)Origin processing method impact on the quality of chuanminshen:To polysaccharides and coumarin as an indicator,the best origin processing methods are 100? boiled with original peel and 70? boiled with original peel.From the perspective of economic energy to consider,choose the 70? boiled with original peel.(2)Experimental Study on the Quality Characteristics of chuanminshen:? Basic quality characteristics:The results showed that moisture content shall not exceed 11%,the total ash shall not exceed 3.5%,acid insoluble ash shall not exceed 0.5%;Water extract and alcohol extract content shall not be less than 30.0%and 5.0%;Crude polysaccharide content of 21.0%?48.0%;? Crude polysaccharide:Crude polysaccharide content of 21.0%?48.0%;The distribution of the different parts of the root structure was:the internal fixation(34.65 ?1.96%)?the posterior column(28.20±1.11%)>the outer skin(10.79± 0.74%);?Coumarin:The average content of isoeperin and isoimperatorin are respectively 0.0097mg/g-0.2687mg/g and 0.0036mg/g?0.0276 mg/g;? Comparison of germplasm resources:Changes of polysaccharide content is wide leaf type(37.80±0.65%)>fine leaf type(20.62±0.07)>lobular type(18.80±0.43);Changes of coumarin content to isoeperin as an indicator is wide leaf type(0.0906mg/g)>fine leaf type(0.0792 mg/g)>lobular type(0.0472 mg/g),the result show that wide leaf type of chuanminshe is high quality resources.On this basis,developed a wide leaf type of Chuanmingshen quality standards for medicines and Pieces(draft)technology and quality control system.4?.Established the characteristic spectrum of TLC?IR and HPLC,and clustering analysis for the characteristics of different components from different origin,established a perfect quality control system of chuanmingshen.It provides an important scientific basis for the study on the comprehensive development and utilization of chuanmingshen. |
