Selenium is one of the essential trace elements of the human body, and it is closely related to human health. In recent years, along with the development of medicine, people pay much attention on the study of selenium. As a trace element essential to the human body, its extensive research and application are involved in food products, medical care, the environment and several other aspects.The edible fungus is delicious, rich in protein, polysaccharide, vitamins, fatty acids and nucleotides. It has become very popular food nowadays. It is not only nutritious, but also has some medicinal value for health care. It shows that the industrialized development of edible fungus is prosperous in the future.Several methods are applied in the selenium content determination, such as atomic absorption spectrophometry, electrochemicalmethods, inductively coupled atomicemission spectrometry, fluorescence spectrophotometry, and spectrophotometry. Among with all these methods, atomic fluores (AFS) has the most important advantages: high sensitivity, low detection limit, high atomization efficiency, wide linear range, no interfere with the matrix, low running cost etc.Objective:1.This thesis is the use of AFS on its characteristics, discussing the quantitative detection and analysis about the selenium content in edible fungus.2. The aim for the thsis is to find the distribution of the selenium content in the edible fungus, provide useful reference on cultivating enriched selenium edible fungus and develope enriched selenium food.3.All these studies will provide invaluable materials for the further research on the relationship of selenium content determination and its distribution & application. Methods:In the practical application of atomic fluorescence analysis technology, the sample pre-treatment technology has also become one of the most crucial techniques. The self-digestion of the sample bottle is used to increase the efficiency of digestion, the entire analysis of theprocess becomes simple and direct. Experiment with other methods of selenium content determinationResults:1.The condition of the selenium determination is optimized, when selected the best equipment and best reaction condition.2.The linear relationship of selenium is in 0~32.0 ng / mL, the correlation coefficient r = 0.9999 for the regression equation Y = 91.328X + 9.208. Determination of blank solution for 11 times, three times the standard deviation divided by the gap in the standard method of calculating the slope of the curve and the detection limit is 0.1201ng/mL. Determination of the 4.00 ng / mL standard solution for 11 times in a row for the fluorescent determination of the value of standard deviation divided by the average measurement, precision reaches to 1.2%.3.The sample recovery is in the 93%~102%, and experimental design achieves the desired purpose.4.The results showed that the selenium content of edible fungi has some relation with the selenium content of the earth and the kind of Edible fungus. Conclusion:AFS is shown to be one of the most effective methods in the determination of selenium content in this thesis. The research proves that the selenium content of edible fungi is effected by the selenium content of the land and the types of edible fungi, provides a theoretical support for the selenium rich edible fungus cultivation. It is hopefully that this technology to test selenium-rich food selection will guide to the future research work. In order to find a solution edible fungus in the lower bioavailability of selenium, it provides a theoretical support for cultivating selenium rich food in order to make a contribution to alleviate or even destroy the eradication of endemic selenium.
