IgA nephropathy is the one of glomerular nephritises that not associated with a group of systemic diseases,renal immunopathology check mesangial area in the existence of a mainly granular IgA deposition,with or without other immune complex deposition,with clinically hematuria or proteinuria with hematuria as the main performance.At present,there is no certainty for its treatment.Professor Deng Tietao' s clinical experience with the Kummerowia striata in the treatment of chronic nephritis hematuria has a good efficacy.This academic dissertation summarizes the status of IgA nephropathy and IgA nephropathy Animal Model Research.The pathogenesis of IgA nephropathy is a complex.On the one hand,increased serum IgA concentration and abnormal glycosylation of mesangial receptor increased with the combination of power, leading to mesangial IgA deposition,is the main incentive for IgA nephropathy; On the other hand,gene polymorphism with IgA nephropathy and the incidence of disease is closely related,suggesting that genetic factors in IgA nephropathy can not be ignored.At present,the pathogenesis of IgA nephropathy is also a lack of consensus,generally believed that,IgA nephropathy pathogenesis of the associated factors include:environmental factors, genetic factors,IgA molecular abnormalities,different regulatory factors such as inflammation.At present,the research focus on the immune,genetic and other aspects.This research through study the general pharmaco-effect of the Kummerowia striata,to explore its pharmacodynamic basic of treatment of IgA nephropathy; through study Kummerowia striata for the treatment of IgA nephropathy,to explore its mechanism that treatment of IgA nephropathy.Through xylene ear edema model in mice,capillary permeability experiments,granuloma model in rats,mice writhing method,hot-plate method to study the anti-inflammatory and analgesic effects;using oslides method,capillary method,study in mice of its coagulation,bleeding time and its impact on platelet aggregation;using experimental mouse ear microcirculation and hemorheology in rat experiments to study the impact of its role of Kummerowia striata;Using metabolic cage method,filter paper observed large load of water animals and acute nephritis in mice of urination function;Using carbon clearance method,the chicken red blood cell hemolysin immunogen for antibody assay,DiNitroToluene (DNCB)-induced delayed-type skin allergic reaction in mice,to observe its effect on immune system.Experimental results show that Kummerowia striata has good anti-inflammatory and analgesic effects(P<0.01);good effect to stop bleeding (P<0.01);with the improvement of microcirculation and blood viscosity changes in the efficacy(P<0.01);a certain degree of diuretic effect(P<0.01); for non-specific immunity,cellular immunity,humoral immunity with the inhibition(P<0.01).This research use BSA+LPS+CCl4 successful preparation of rat model of IgA nephropathy,to observe rats normal circumstances,urine routine examination, blood biochemical,pathological,blood and kidney SOD vitality and MDA content,and to study Kummerowia striata' s effect on IgA nephropathy and its mechanism.The results show that the Kummerowia striata could significantly improve the lives of rats,and reduce the amount of proteinuria and hematuria,improving renal function and slow down the kidney mesangial proliferative.IgA nephropathy model rats' SOD vitality descended,MDA content increased in vivo that decreased antioxidant capacity,oxygen free radicals have a number of enhanced,lipid peroxidation enhancement, Kummerowia striata treatment group can Increased SOD vitality and lower MDA content,reduce the symptoms of IgA nephropathy,the prevention of disease development.Prompted that Kummerowia striata has a clear free radicals and antioxidant enzymes to enhance vitality,reduce or block the lipid peroxidation chain reaction the role of treatment may also be one of the mechanisms of IgA nephropathy.
