Objective:Tupistra Chinensis Bak. belongs to Liliaceae species. Chinese traditional medicine theory considers that this plant has the effect of heat-clearing and detoxicating, expel wind-evil and remove wetness, eliminating stasis to activate blood circulation and odynolysis. Tupistra Chinensis Bak. is commonly used in the treatment of pharyngitis, tonsillitis in the civil, the effect of which is significant. In recent years the article reported that Tupistra Chinensis Bak. has effect of anti-inflammatory and expectorant. This study aims to investigate and verify whether the Tupistra Chinensis Bak. has the effect of antitussive, expectorant, antiasthmatic and therapeutical effects to Chronic Bronchitis through in vivo and in vitro experiment.Method:1 Antitussive and expectorant effects of TCE1) Antitussive effect of TCETussis of strong ammonia irritation was used. The incubation period of cough and times in 3 minutes were recorded from beginning ammonia irritation to presenting cough. Results showed that Low dosage group of Tupistra Chinensis Bak. Extract(L group) and High dosage group of Tupistra Chinensis Bak. Extract(H group) compared with Model group(M group), the delitescence period of cough were prolonged obviously (P<0.05,0.01), the cough times reduce obviously (P<0.05,0.01). The incubation period of cough and the cough times of Dissolvant group were equal to M group(P>0.05).2) Expectorant effect of TCETracheal phenol red secretion was used. Phenol red secretions in every groups were calculated. Results showed that L group compared with M group, tracheal phenol red secretion increase insignificantly(P>0.05);tracheal phenol red secretion on H group compared with M group increase obviously(P<0.05). Tracheal phenol red secretion of dissolvant group were equal to M group(P>0.05).2 Anti-inflammatory and tracheal smooth muscle relaxation effect of TCE 1) Anti-inflammatory effectXylene-induced ear swelling in mice was used to calculate the swelling inhibition rate by the degree of ear swelling which based on the difference between the left and right ear. Results showed that L group and H group compared with M group, the degree of ear swelling decreased obviously(P<0.05,0.01), and the swelling inhibition rates were 46.04 percent, 60.99percent. The degree of ear swelling of dissolvant group were equal to M group(P>0.05).2) Tracheal smooth muscle relaxation effects of TCEThe guinea-pig isolated trachea spiral strip was used to evaluate the role of TCE on the guinea-pig isolated trachea spiral strips. Results showed that TCE had no inhibitory effects on the contraction of the guinea-pig isolated trachea spiral strip induced by Ach (P>0.05), and no contribution to the concentration-response curves of Ach (P>0.05).3 Effects of TCE on the serum TNF-α, IL-6 of chronic bronchitis mice and Pathological Changes of pneumono-tissueFourty-eight mice were divided into six group: the normal group(N group), the model group(M group), the anti-inflammatory tablets control group(A group), the high-dose of TCE group(H group), the low-dose of TCE group(L group), the dissolvent control group(D group). Exposure to cigarette-smoking was used to establish the mice model of chronic bronchitis. ELISA kit were used to determinate the content of serum TNF-α, IL-6 as well as the pathological changes of pneumono-tissue.Results showed that①The content of serum TNF-α, IL-6 of M group increase obviously (P<0.01) compared with B group. A group, H group, L group significantly decreased (P<0.01) compared with M group.②P athological Changes of pneumono-tissue: N group: The construction of bronchus and the epithelium of tunica mucosa bronchiorum is normal, no inflammatory infiltration, no exudation in tracheal cavity. M group: Endothelial cells of tunica mucosa bronchiorum seriously necrosis, mucilaginous glands accrementition, basilemma thickening, liquid exudation, bronchus infiltrate with large amount of inflammatory cells, circum- bronchus congestion, dropsy, vessel expansion. A group: The lung bellows is basic-normal, endothelial cells of tunica mucosa bronchiorum complete, the tube has no the exudation, sometimes the caliciform cell proliferate with the inflamed cells. H group: Pathological Changes of lung morphology and bronchial epidermis damage improved remarkably, accrementition of basilemma and exudation in tracheal cavity decrease, the inflammatory cell of the bronchus and lung tissues reduce obviously. L group: the bronchial epidermis damage are improved remarkably, infiltration with inflammatory cells and the mucous membrane proliferation decrease, endothelial cells of tunica mucosa bronchiorum complete, the secretion reduces.Conclusion:1 TCE displayed the antitussive and expectorant effects.2 TCE obviously showed the anti-inflammatory effect but had no inhibitory effects on the guinea-pig isolated trachea spiral strip .3 TCE reduced the content of serum TNF-α, IL-6 and the inflammatory cells infiltration around the lung bellows, improved the construction of bronchus and pathology morphological changes of the lung bellows.As a conclusion, TCE could be further developed to became a new type of Chinese medicine for the treatment of respiratory diseases because of it's antitussive, expectorant and Anti-inflammatory effect.
