Tupistra chinensis Baker is belongs to Convallarieae of Liliaceae family, whose saponin extracted from the dry rhizome of plant. As the usual medicinal materials of Shennongjia Natural Reservation Area, Tupistra chinensis Baker is frequently used to treatment on some diseases, for instance, headache, cough, sore throat and so on. Due to the limitation of resource distribution, there are not systematic studies on the extraction method and pharmacological action about anti-atherosclerosis of the Tupistra chinensis Baker saponins. Our team researched on the effect and mechanism about anti-hyperlipidemia and anti-atherosclerosis of the saponins, by optimizing the extraction technology of the Tupistra chinensis Baker saponins.In order to determine the optimum extraction process of the Tupistra chinensis Baker saponins, heating reflux method, ultrasonic method, microwave method and supercritical fluid extraction were discussed in this paper. Employed orthogonal experimental design to examine the influencing factors for the extraction, with the use of the extraction rate of the total saponins of Tupistra chinensis Baker as the evaluation index.The optimized extraction technology was as follows: the samples were subjected to reflux extraction for 3 times with 10 times amount of 70% ethanol at 90℃, each extraction lasting 1h. Meanwhile, we used the ultrasonic method and microwave method to extract the plant saponin on the base of ethanol extraction.The extraction rate is respectively 3.154%and 3.482%, lower than the heating reflux method's. In addition, The extraction rate of supercritical fluid extraction is 5.822%, much more higher than the first three methods. The experimental results have shown that, as an extraction method possesses the advantages of normal temperature, low-pollution and high-yield, the supercritical fluid extraction should play an important role in industrial production.To observe the effect of saponin extracted from Tupistra chinensis Baker on blood lipid metabolism and the inhibition of cholesterol absorption in experimental hyperlipidemia rats, we tested the effects on blood-fat biochemical indicator, the inhibition of cholesterol indexmicrozyme and the lesions of aorta in the rat models by establishing several groups, blank control, model, positive control drug and drug control group. Compared with model group, it was found that TC, LDL-c, TC/HDL-c and LDL-c/HDL-c were decreased significantly and HDL-c were significantly increased by given different dose of saponin in experimental hyperlipidemia rats.; high-dose group could relieve the symptom of AS; Besides, the saponin could inhibit the formation of cholesterol indexmicrozyme in the intestinal tract to some extent. Consequently, Tupistra chinensis Baker saponins can heal experimental hyperlipidemia; inhibit the formation of cholesterol indexmicrozyme and atherosclerotic plaque, provide evidence for clinical therapy of hypertension, angina pectoris and atherosclerosis.
