| Objective:Type 2 diabetes is an autoimmune disease characterized by T-cell mediated autoimmune destruction of pancreaticβ-cells.It's pathogeny due to a combination of genetic and environmental factors.Infection of Coxsackie B virus is one of the major risk factors.Immune mechanism plays a main role in the occurring of type 2 diabetes.The aims of this study were to examine the CVB-IgM(CVB3-IgM,CVB4-IgM,CVB5-IgM) in sera and cell factors including interleukin-1β,interleukin-6, interleukin-10,interleukin-12,interleukin-18,tumor necrosis factors-α,IFN-γin sera with Type 2 diabetes,and to investigate the relation between infection of Coxsackievirus group B and type 2 diabetes with Th1/Th2 imbalance.This study will provide valuable guidance for type 2 diabetes treatment and prevention.Method:Type 2 diabetes was diagnosed based on the standard of WHO.The CVB-IgM(CVB3-IgM.CVB4-IgM.CVB5-IgM)in sera and cell factors including interleukin-1β,interleukin-6,interleukin-10,interleukin-12,interleukin-18,tumor necrosis factors-αin sera with 300 patients(162males,138females) and 300 normal(156males,144females) controls were detected by ELISA.Results:1.6.7%of the patients and in 1%of normal control group p<0.01.The proportion of CVB3-IgM,CVB4-gM,CVB5-IgM in sera are 10%,65%,and 25%respectively.2.The levels of IL-1β,IL-6,IL-10,IL-12,IL-18,TNF-α,IFN-γin patients were higher than that in controls.There are significant differences in IL-1β,IL-6,IL-18,TNF-α,IFN-γ.(P<0.05)3.The levels of IL-1β,IL-6,IL-10,IL-12,IL-18,TNF-α,IFN-γin type 2 diabetic patients with positive CVB-IgM were higher than those with negative. There are significant differences in IL-1β,IL-6,IL-18,TNF-α,IFN-γ(P<0.05) 4.Plasma IL-1β,IL-6,IL-10,IL-12,IL-18,TNF-αin 20 patients were detected in pretherapy and post-treatment by insulin.The leves of IL-1β,IL-6,IL-10,IL -12,IL-18,TNF-α,IFN-γin post-treatment group were lower than that of pretherapy group.There are significant differences in IL-1β,IL-6,IL-18,TNF-α,IFN-γ(P<0.05)Conclusion:The results suggested that Type 2 diabetes and infection of Coxsackievirus group B was correlated.Viurs-mediated autoimmune disorder makes T-cell activation.The increase of IL-1β,IL-6,IL-10,IL-12,IL-18,TNF-α,IFN-γsecretion.Cell factors activate several pathways of apoptosis and promote theβ-cell apoptosis.CVB infection is associated with T-cell mediated immunity in Type 2 diabetic patients. |
