Effects Of Celery On Sperm Quality And Fertility Of Male Mice

Objective to explore the effect of celery on sperm quality and fertility of male mice, so as to provide the basic scientific materials for the utilization of celery.Methods 240 adult male mice Of Kunming strain in SPF degree were randomly divided into 2 experimental groups according to the body weight including acute experimental group,subacute experimental group.Each experimental group were randomly divided into 4 subgroups(normal saline group,the low-dose group,the medium-dose group and high-dose group),and there are 30 male mice in each Subgroup.Mice of each group were administered by gavage at a certain time every day,experimental group with 0.3ml of celery juice of different concentration and control group with 0.3ml of normal saline.(1)10 mice of each subgoup were randomly selected and killed by dislocation respectively at the end of 7 days(acute experimental group) and 56 days(subacute experimental group),then sperm density,sperm activate rate and sperm motility parameters were measured by WLJY-9000 color-detection system of sperm quality.(2) At the end of intragastric administration,10 mice of each subgoup were randomly selected and were mated 1:1 with untreated females for 7 days(time for completion of one estrous cycle in mice.The daily vaginal smears were checked at 8.00Am everyday.The day that vaginal plug was found was considered day 0 of gestation.Then male mice were killed at the end of mate for 7 days,sperm quality parameters were measured.Mice were killed by dislocation and checked by caesarean on day 18 of gestation.The pregnant rate,embryonic nidation of each group were recorded,and the index including mating index,fertility index, pregnancy rate were calculated.(3) Remaining mice were killed after recoverd for 2 weeks,and sperm quality were detected. Results(1) The mice of two experimental groups had good growing developments after intragastric administration of celery juice,the weights of all mice were all increasd(P＞0.05).(2)In gavage groups of subacute experimental groups liver index of the medium dose group and high dose group were significantly increased,on the basis of excluding the effect of weight change on the viscera coefficient change.(3)The hemoglobin of two experimental groups Had not obvious variations(P＞0.05).(4) In medium dose group of acute experiment,the sperm density was increased,the proportion of sperm which was c degree(non-forward moment) and d degree(no move) was raised,sperm motility and sperm activate rate were significantly decreased,perm motility parameters were not significant changed;sperm quality of mated group and recovered group was not obvious Variations,which did not show significant difference compared with inter blocks.(P＞0.05)(5)The effect of mice serum hormone was not detected,which did not show significant difference compared with every group(P＞0.05).(6) In the acute experiment and subacute experiment,fertility index,mating index of gavage group were decreased disproportionately,which did not show significant difference compared with control group(P＞0.05).The Number of live of nest,average weight and average length had not statistic significance between every dose group (P＞0.05).Conclusion Celery had do effect on sperm quality in some degree.,Celery did not have some obvious effect on the growing development of rat fetus,but the effect on the fertility had no been excluded.
