Objective The expression of wild-type p53-induced phosphatase 1(Wip1)gene in human sperm was studied and its correlation with sperm concentration and viability was investigated.Methods The male subject clinic of the First Affiliated Hospital of Guangxi Medical University Semen samples of normal semen,oligozoospermia,weak sperm disease,and oligozoospermia were collected,and 20 samples in each group.Then the sperm DNA fragmentation index(DFI)was detected by sperm chromatin diffusion method(SCD method),the sperm survival rate of each group was detected by rejection method,and the spermatozoa of each group were detected by real-time fluorescent quantitative PCR(q-PCR)and immunofluorescence.The expression of Wip1 was analyzed and its expression characteristics were analyzed.Results(1)There were significant differences between the oligozoospermia group,the weak spermatozoa group and the oligospermia group compared with the normal group(P<0.05).There was no significant difference in sperm survival rate between the oligozoospermia group and the normal group(P>0.05),the survival rate of the weak spermatozoa group and theoligospermia group were significantly lower than those of the normal group(P<0.05);(2)The difference between survival rate and viability between DFI>25% and DFI≤25% was statistically significant(P<0.05),but there was no significant difference in sperm concentration between the two groups(P>0.05);(3)The results of q-PCR showed that there was no significant difference in Wip1 mRNA expression between the normal group and the oligozoospermia group(P>0.05),while the weak spermatozoa group and the oligospermia group had statistical significance compared with the normal group(P< 0.05),negatively correlated with sperm motility(r=-0.412,P<0.05),positively correlated with sperm concentration [replaced: positive correlation](r=0.321,P<0.05),and positively correlated with DFI(r=0.306,P<0.05);(4)Compared with the results of immunofluorescence in the 4 groups,the expression of Wip1 protein in the normal group and the oligozoospermia group was not statistically significant(P>0.05),and the expression of Wip1 protein in the weak spermatozoa group and the oligospermia group was higher than that in the normal control group.(P<0.05).Conclusion It is confirmed by experiments that Wip1 is expressed in human sperm.There is a positive correlation between Wip1 expression and sperm DFI.After sperm DNA damage,the expression of Wip1 is increased,which may negatively regulate sperm DNA damage repair process,resulting in decreased sperm motility.It is also possible to dephosphorylate its downstream pro-apoptotic targets,thereby affecting sperm concentration.
