ObjectiveTo supply the scientific experimental foundation for spleen-kidney re lativity and to explore the mechanism of action of Qiangji Jianli(QJJL) F ang for treatment of spleen-kidney deficiency syndrome by creating a mode l of spleen-kidney deficiency syndrome,and at the same time use drugs s uch as QJJL to protect and treat.MethodsTheoretical study:An overview of the past few years about the QJJL, Huang Qi,and spleen- kidney relativity were given firstly.According to the overview,we planed to create a model of spleen-kidney deficiency by gavage with rhubarb followed with hydrocortisone,and select the past re ported indications which have a better reproducibility to observe,such a s Thyroid hormones,sex hormones.Experimental study:choose the healthy SD male rat to the Experimenta l animal.We did our experiments by two steps.Experimentâ… Effect of Qian gji Jianli Fang on the Contents of hormonein Rat Models of Spleen-Kidney Deficiency Syndrome.We created a model of spleen-kidney deficiency by gavage with rhubarb followed with hydrocortisone.During creating the model,we used the med icine to protect and treat.There were three drug groups to design:the QJJ L group,the Qiji Yougui(QJYG) wan group,the Yougui Wan(YGW).We applie d QJJL in the QJJL group during creating the model;applied YGW in the YG W group;in the QJYG wan group,applied QJJL in spleen and YGW in kidney. We selected the indications such as thyroid hormones,sex hormones,adre nocorticotropic hormone to observe.So we try to explore the spleen-kidne y relativity and the mechanism of action of QJJL Fang from experimentâ… by observing the change of these indications in each group. Experimentâ…¡The Research about primary Effect of Huangqi on QJJLThis experiment was the basis of experimentâ… .we created the same ani mal model and selected the same indexes to observe.there were three drug groups:the QJJL group,the Huangqi Jianliang(HQJL) Fang group,and the Huangqi Danfang(HQDF) group.We try to explore the mechanism of action of Huangqi in the QJJL fang by observing the different condition in each group.ResultsExperimentâ… :Compared with the normal group,The changes of the temp erature in the model group was lower in the spleen deficiency(P<0.05),a nd more lower in the spleen-kidney deficiency(P<0.01).Compared with the model group,the temperature in the QJJL,and QJYG group was higher(P<0. 05-0.01).Compared with the normal group,the T3,T4,rT3 content was poorer in t he spleen and spleen-kidney deficiency(P<0.01),TSH did not change signif icantly.Compared with the model group,the T3,T4,rT3 content of the QJJL, QJYG and YGW groups increased in the spleen deficiency(P<0.01);the eff ectof each group were still in the spleen-kidney deficiency but YGW to rT3 content.Compared with the normal group,the content of T rose firstly but fell significantly later(P<0.01).The content of E2 in the spleen deficiency a nd spleen-kidney was higher significantly(P<0.01).E2/T ratio increased s ignificantly in the spleen-kidney deficiency(P<0.01).Compared with the model group,the T content of the QJYG group decreased in the spleen defi ciency(P<0.05),and increased in the spleen- kidney deficiency(P<0.01). The E2 content,and E2/T ratio in each group fell in the spleen- kidney d eficiency(P<0.05-0.01).In the spleen deficiency and the spleen-kidney deficiency,the conten t of ACTH was higher(P<0.05-0.01).Compared with the model group,the AC TH content of the YGW group decreased in the spleen-kidney deficiency(P< 0.05).Compared with the normal group,content of cAMP,cAMP/cGMP ratio were higher during the spleen deficiency and the spleen-kidney period(P<0.05). The content of cGMP has no change significantly firstly,but declined lat er(P<0.05).Compared with the model group,cAMP content,cAMP/cGMP ratio of the QJYG group were lower in the spleen deficiency(P<0.01).In the spl een-kidney deficiency,cAMP content,cAMP/cGMP ratio fell and The content of cGMP increased in each groups(P<0.05-0.01).Experimentâ…¡Compared with the normal group,the body weight lessen apparently in the spleen and the spleen-kidney deficiency(P<0.01).Compared with the mo del group,the QJJL,HQJL,and HQDF can increase the body weight(P<0.05-0. 01).Compared with the normal group,the T3,T4,content was poorer in the s pleen and spleen-kidney deficiency(P<0.01),TSH did not change significan tly.rT3 content increased in the spleen deficiency(P<0.05)and didn' t ch ange in the spleen-kidney deficiency.Compared with the model group,theT 3,T4 content of the QJJL,HQJL and HQDF groups increased in the spleen and spleen-kidney deficiency(P<0.01).In the spleen deficiency,the QJJL and HQDF can decrease the content of rT3(P<0.05).Compared with the normal group,the content of T didn' t change firstl y but fell significantly later(P<0.05).The content of E2 and E2/T ratio i n the spleen and spleen-kidney deficiency was higher significantly(P<0.0 1).Compared with the model group,the T content of the QJJL and HQDF gro up increased and the E2 content,and E2/T ratio fell in the spleen- kidney deficiency(P<0.01).There were not changes in the spleen about the content of ACTH and Co r.In the spleen-kidney deficiency,the content of ACTH was higher,and C or was poorer(P<0.01).Compared with the model group,the ACTH content of the YGW group decreased in the spleen-kidney deficiency(P<0.01).Compared with the normal group,content of cAMP,cAMP/cGMP ratio were higher(P<0.01),and the content of cGMP declined during the spleen defici ency and the spleen-kidney period(P<0.01).Compared with the model group, cAMP content,cAMP/cGMP ratio of the QJJL,HQJL,HQDF group were lower(P<0. 05-0.01),and the cGMP content of the HQJL,HQDF group were higher in the spleen deficiency(P<0.01).In the spleen-kidney deficiency,cAMP content, cAMP/cGMP ratio fell(P<0.05) and The content of cGMP increased in QJJL groups(P<0.01).Compared with the normal group,the content of AMS was lower significa ntly in the spleen deficiency(P<0.01),but not change in the spleen-kidne ydeficiency.Compared with the model group,the AMS content of the QJJL gr oup was higher in the spleen deficiency(P<0.01).There were not changes significantly in each group about SOD,MDA. Pathological changes were found in pituitary,thymus,spleen,kidney,and adrenal.The expression of thymus PCNA protein decreased in the spleen-ki dney deficiency(P<0.01).The QJJL can protect these organa and increased the expression of thymus PCNA protein(P<0.01).ConclusionThere was a link between the spleen and kidney,not only in mutual pa thological changes,but also in the treatment of each other.It may be th e one of the main channel for spleen-kidney contact the hypothalamus-pitu itary-target gland axis.There were upper change in the kidney deficiency, and lower changes in the spleen deficiency.By observing the change in t he hormone content,we can have a preliminary understanding of the pathol ogical of spleen and kidney,as well as the effect of treatment.QJJL can effectively prevent and treat the spleen deficiency and the spleen-kidney. Large dosages of huangqi play an important role in QJJL.If we decrease the dosage of huangqi or a singal use of huangqi,the effective of QJJL wo uld reduce or disappear.The mechanism may be that huangqi can directly a nd indirectly tonify the kidney.combined with other drugs,the ingredien ts of QJJL is easier to reach the target organ to play a role.
