Study On The Relationship Between Single Nueleotide Polymorphisms Of HSP70 And JWA Genes And Susceptibility To Hypertension Of Workers Exposed To Heat Stress

Objective: To protect occupational population from hypertension and to study on hypertension influencing factors of workers exposed to heat stress. To study the relationship between HSP70 and JWA genes polymorphisms and hypertension of workers exposed to heat stress. To study on the feasibility of HSP70 and JWA genes polymorphisms as biological marker of hypertension susceptibility.Methods: 160 steel workers and rollers were selected in the exposure groups. 114 workers unexposed heat stress were selected in the control groups. We investigated their general information and the work environment. All subjects accepted health examination including anthropometric characteristics (body heigh,tweight,waistline),blood pressure,blood glucose,triglyceride and cholesterin. HSP70-1 gene 190 site,HSP70-2 gene 1267 site,HSP70-hom gene 2437 site,JWA gene 76 site and JWA gene 723 site genotypes were analyzed by using the polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)technique. Using PHASE 2.0 software to calculate haplotype.Results: The prevalence rates of hypertension in the exposure groups and control groups were 25.00% and 12.28%. The rate was significantly higher in the in the exposure groups (P<0.05). Through multiple logistic regression analysis, We found that high temperature,obesityand smoking were risk factors for hypertension, obesity was a risk factor in non-exposure groups(OR=19.330, P<0.05),smoking and cholesterin were risk factors in exposure groups (OR=16.288,2.989,P<0.05).The exposure groups of<35,35~45 and >45 were 29.03%,21.33% and 26.08% in hypertension morbidity that were larger than non-exposure groups, respectively. There was significant difference in these two groups: the exposure groups of 35~45 and non-exposure groups (P<0.05).HSP70-1,HSP70-2 and HSP70-hom genotype frequencies of the exposure groups and the non- exposure groups were no significance differernces in normal blood pressure,blood pressure higher population and hypertension. HSP70-1 C/C genotype frequencies were 23.52%,36.36% and 15.00% in the normal blood pressure,blood pressure higher population and hypertension of the exposure groups. In the non-exposure groups, they were13.73%,38.46%,16.67%. In all subjects, they were19.85%,36.96%,15.38%. HSP70-1 C/C genotype frequencies of blood pressure higher population were higher than that of the normal blood pressure and hypertension of the exposure groups and the non- exposure groups. HSP70-1 C/C genotype frequencies of blood pressure higher population were significantly higher than that in hypertension of the exposure groups and all subjects. HSP70-2 G/A genotype frequency in the blood pressure higher of the exposure groups was significantly lower than that in hypertension and normal population of the exposure groups (P<0.05). Through multiple logistic regression analysis, we found that the G/A genotype in the exposure groups was a protective factor for blood pressure elevation, OR value was0.39 (95%CI: 0.16~0.95), adjustment OR value was0.25 (95%CI: 0.08~0.78). We found that the GAT/***+ ***/***haplotype pairs were risk factors for hypertension of the exposure groups, OR value was2.43 (95%CI: 1.03~5.35), adjustment OR value was2.00 (95%CI: 0.80~6.77).JWA76 G/G genotype frequencies were 82.35%,69.70% and 65.00% in the normal blood pressure,blood pressure higher population and hypertension of the exposure groups. JWA76 G/C genotype frequencies were 17.65%,30.30% and 35.00% in the normal blood pressure,blood pressure higher population and hypertension of the exposure groups. There was significant difference in the normal blood pressure,blood pressure higher population and hypertension of the exposure groups of JWA76 G/G and G/C genotype frequencies (P<0.05). Through tenfency analysis, we found JWA76 G/G genotype frequencies decreased gradually with blood pressure elevation, JWA76 G/C genotype frequencies increased gradually with blood pressure elevation(χ2=4.86,P=0.027). Through multiple logistic regression analysis, we found that the G/C genotype was risk factors for hypertension of the exposure groups, OR value was2.51 (95%CI: 1.09~5.91), adjustment OR value was3.67 (95%CI: 1.21~11.05). The GG/CT was risk factors for hypertension of the exposure groups, OR value was3.39 (95%CI: 1.01~11.46), adjustment OR value was8.55 (95%CI: 1.53~47.48).Conclusion: 1. Heat induced hypertension, it induced age ahead of schedule. Smoking and cholesterin were cooperation factors in hypertension development.2. The gene polymorphism at HSP70-2 gene 1267 site had association with blood pressure elevation and GAT/*** +***/***haplotype pairs had association with hypertension. HSP70-1 C/C genotypes have association with blood pressure elevation.3. The gene polymorphism at JWA gene 76 site and GG/CT haplotype pairs had association with hypertension.
