In this article, the content of 5-HT and the key rate-limiting enzyme were assayed by chemiluminescent fluorescence and electrophoresis experiment separately. The sleep efficiency to be used the royal jelly and gastrodia elata B1. were researched. The results are as follows:1. In this article, pharmacodynamic experiment of sleep-improvement was carried out by feeding mice on royal jelly with different concentration. Sleep quality of mice is judged by the sleeping incubation period time of the pentobarbital sodium, the sleeping extended time of the pentobarbital sodium and the rate of sleeping induced by subthreshold-dose of the pentobarbital sodium. The results showed that the doses of sleep-improvement were 0.208 g/mL, 0.313 g/mL and 0.417 g/mL separately.2. The doses of gastrodia elata B1. which improved sleeping were determined by the same methods and judgment index with the royal jelly. The results showed that the doses of sleep-improvement were 0.500 g/mL, 0.625 g/mL and 0.750 g/mL separately.3. The royal jelly with three different doses of the gastrodia elata B1. separately were combined. and the best dose of the compound was determined by used the same methods and judgment index.Then the compound which have got was feeded on mice. In the mice brain the content of 5-HT and the key rate-limiting enzyme were assayed by chemiluminescent fluorescence and electrophoresis experiment separately . The results showed that when combined the royal jelly with gastrodia elata B1. by 1:2, the effect of improving sleep was the best. Compared with the other groups, the compound prescription may clearly shorten the sleeping incubation period; and it can also extend the sleeping time of the pentobarbital sodium; at the same time it can clearly increase the rate of sleeping which induced by subthreshold-dose of the pentobarbital sodium. The effect of the compound on the mice showed that the best compound prescription had no side effects. The result of the content of 5-HT in the brain of mice showed that the best compound prescription can increase the content of 5-HT; and the result of the key rate-limiting enzyme showed that the best compound prescription did not change the content of tryptophan hydroxylase in the brain of mice.In summary, when combined the royal jelly with gastrodia elata B1. by 1:2, it was beneficial for improving sleep of mice.
