Objective:Systemic lupus erythematosus(SLE) is a kind of autoimmunedisease caused by many factors accompany with multi organs damage,the etiological factor refer to heredity,environment,abnormal of immunoloregulation,sexual hormone and so on.To study the mechanism of estrogen receptors and T,B leukomonocyte synergistic effect in SLE,we approach through the experiment:1.the expression difference of estrogen receptor-α(ER-α),interleukin-10(IL-10),B lymphocyte stimulating factor(Blys) in health adult and SLE patients of male and female;2.the correlation between the mRNA expression of estrogen receptor-α,interleukin-10,B lymphocyte stimulating factor and clinical and laboratory indexes;3.the correlation between the mRNA expression of ER-αand IL-10,Blys,to approach ER-α's effect in T and B leukomonocyte,and the synergistic effect in T and B leukomonocyte of SLE patients.To offer some theory basis in elucidating pathogenetic,monitoring reactiveness,drug exploitation of SLE.Method:RT-PCR method is used to analysis the expression quantity of ER-α, IL-10,Blys in SLE patients(40 cases) and health adults(40 cases),The PCR products are separated by electrophoretic method through gelose and ethidium bromide which concentration is 4.0%.The consequence is judged using gel formatter and statistical analysis.According to the diagnostic criteria and analyze the expression difference of ER-αand IL-10,Blys and the correlation between the mRNA expression of ER-αand IL-10,Blys using the statistical software,approach ER-α's effect in T and B leukomonocyte,and the effect in T and B leukomonocyte of SLE patients.Result:1.The mRNA expression level of ER-αand IL-10,Blys in PBMC of SLE patients is obviously higher than normal control(P<0.05 or P<0.01).2.The mRNA expression level of ER-αand IL-10,Blys in PBMC in active group is obviously higher than inactive group(P<0.05 or P<0.01). 3.The mRNA expression level of ER-αand IL-10,Blys in PBMC of SLE patients have no statistical significance between male and female in SLE group and control group(P>0.05).4.The expression level of ER-αwith kidney damage is higher than the non- kidney damage group,show a positive correlation with ESR,24 hours quantitation of urine protein(P<0.05),and show a negative correlation with C3(P<0.01).There has no correlation wih the other indexes,for example fever,erythra,alopecie,ANA,Sm and so on(P>0.05).5.The expression level of IL-10 with arthritis and kidney damage is higher than the non-damage group,show a positive correlation with IgG and SLEDAI(P<0.05 or P<0.01),and show a negative correlation with C3(P<0.01).6.The expression level of Blys with serous cavity fluidify and kidney damage is higher than the non-damage group,show a positive correlation with ds-DNA titre,IgG and SLEDAI,and show a negative correlation with C3 and C4(P<0.05 or P<0.01).There has no correlation wih the other indexes(P>0.05).7.The mRNA expression level of ER-αshow a positive correlation with IL-10 and Blys,the mRNA expression of IL-10 and Blys also show a positive correlation (P<0.05 or P<0.01).Conclusion:1.The mRNA expression level of ER-αand IL-10,Blys in PBMC of SLE patients is elevated than normal control which suggested that ER-α,IL-10,Blys may take part in the pathology of SLE.2.The mRNA expression level of ER-αand IL-10,Blys in PBMC in active group is obviously higher than inactive group and show a positive correlation with SLEDAI which suggested that ER-αand IL-10,Blys may be related with the reactivity of SLE.3.The mRNA expression level of ER-αin PBMC have a correlation with kidney damage and 24 hours quantitation of urine protein,the mRNA expression level of IL-10,Blys correlate with kidney damage which suggested that they may take part in development of SLE and correlate with the severity of SLE.4.Acconding to the mRNA expression level of ER-α,IL-10 and Blys in PBMC with the clinical and experimental indexs,we can conclude that ER-α,IL-10 and Blys take part in immunity reaction and inflammatory reaction,and they may be inflammation and immunity indexs of SLE.5.Acconding to the correlation between the mRNA expression level of ER-αand IL-10,Blys,we conclude that ER-αmay have active effect on IL-10 and Blys in gene level,ER-αmay regulate IL-10 and Blys and enhance the mRNA expression of the two cytokines,ER-αmay have an important role in the pathology of T and B lymphocyte of SLE.6.The experiment show a positive correlation between the expression of IL-10 and Blys which suggest that in the pathology of SLE,T and B lymphocyte may interact with each other to induce SLE.7.The further study of ER,IL-10 and Blys may offer some theory basis in elucidating pathogenetic,monitoring reactiveness,drug exploitation of SLE.
