| Objective To study the effects of Curcumin on the expression of transforming growth factor-β1(TGF-β1),serum and glucocorticoid-inducible kinase1(SGK1),connective tissue growth factor (CTGF) on the process of renal interstitial fibrosis and SGK1 in renal glomerular caused by unilateral ureteral obstruction (UUO) in rats. Methods Fifty-four Sprague-Dawley rats were randomly divided into three groups: sham operated group,UUO model group,Curcumin-treated group.Each group has eighteen rats.The rats in the Curcumin-treated group and UUO model group underwent UUO, rats in sham group had their ureters manipulated but not ligated. Six rats of each group were killed respectively at 3,7,14 days after surgery. The level of BUN and Scr was observed. Histological changes were observed by HE and Masson staining. Immunohistochemistry was also performed on renal interstitial tissue for TGF-β1,SGK1,CTGF and renal glomerular SGK1 protein. Results In the obstructed kidneys, there were significant renal interstieial fibrosis and interstitial space expansion. Immunohistochemical staining showed that the expression of TGF-β1,SGK1 and CTGF was significantly increased in the obstructed kidneys. In comparison with the model group,Curcumin could obviously decrease interstitial volume and the expression of TGF-β1,SGK1,CTGF and renal glomerular SGK1.It also improved the histological changes of the UUO renal tissue. But the level of BUN and Scr was not significantly altered. Conclusions It was revealed that Curcumin can down-regulate the expression of TGF-β1,SGK1 and CTGF in UUO rats. This demonstrated its potential ability to inhibit renal interstitial fibrosis caused by UUO in rats. |
