| The mutagenic and estrogenic effect of the nonvolatile organic compounds (NOCs) ,which was concentrated from the source water and drinking water (plant water and tap water) of the Changjiang and Hanjiang River in Wuhan at calm, high and low flow period, were detected and evaluated by the Salmonella mutagenicity test (Ames test) and Yeast Estrogen Screen (YES). This research will give scientific data about the toxic character, intensity and regularity of the NOCs in water, the adjustment of the water treatment technology of the correlated water plant, the safety of biology and chemistry of the drinking water, the establishing and consummating of the planning, management, exploitation, utilization and protection of the water resources, and the steps about the control of the water pollution instituted by the section such as water conservancy, environmental protection, public health and so on.The source and drinking water (plant water and tap water) of the Changjiang and Hanjiang River was collected in Wuhan in Mar.2007 (calm flow period), Jul.2007 (high flow period) and Dec.2007 (low flow period) successively. The volume of each sample was 100~120L. Simultaneously we collected the water quality parameters of the water samples. The organic compounds in water samples was absorbed by XAD-2 resin and eluted by dichlormethane and acetone sequentially. After evaporated and metered volume, the extracts were detected and compared by Ames test and YES. The NOCs of Each sample was negative to TA100 except plant water of Hanjiang River without S9 mix at calm flow period. The NOCs of each sample without S9 were positive to TA98 except source water of Changjiang River at low flow period and plant water of Changjiang River at high flow period, the mutagenicity of the samples with S9 was lower than which without S9 . The mutagenicity of the NOCs about the water samples at different periods was low flow period > calm flow period > high flow period. All the NOCs of the plant water and tap water samples with or without S9 mix were obviously higher than the source water. The mutagenicity of the NOCs about plant water was greater than tap water at calm and high flow period, but at low flow period tap water was greater than plant water. The mutagenicity of the NOCs about Changjiang River was higher than Hanjiang River at calm and high flow period, but it was lower than Hanjiang River at low flow period. The estrogenic activity of the NOCs about source water of Changjiang and Hanjiang River was higher than plant water and tap water. The estrogenic activity of NOCs about Changjiang River was higher than Hanjiang River. The estrogenic activity of the NOCs about the water samples at different periods was low flow period > calm flow period > high flow period. Conclusion The NOCs of each sample was essentially negative to TA100 and positive to TA98 at the extent of the density, the main mutagen was frameshift type. The source water of Changjiang and Hanjiang River showed estrogenic activity and the intensity of Changjiang River was greater than Hanjiang River at every period. The mutagenicity of the NOCs about Changjiang River was higher than Hanjiang River at calm and high flow period, but lower at low flow period. The mutagenicity of NOCs in water increased after treated of routine process, while the estrogenic activity decreased significantly after treated. The intensity of the mutagenicity and estrogenic activity of the NOCs about the water samples at different hydrographic periods was low flow period > calm flow period > high flow period. |
