The research mainly is that the progesterone's influence to sperm capacitation and acrosome reaction in developing fast ART recently.The rate of fertilization is the essential step for ART success ratio,but the mechanism about progesterone is very little,so the research is chosen to discuss.The purpose is processing the human's sperm with the different concentration progesterone,namely,appending different concentration progesterone into sperm capacitating medium,through observation application of progesterone or not to the influence of the test of sperm binding oocyte's zona pellucida(ZP),and observing sperm capacitation and the acrosome reaction occurrence situation,to supply information for clinical and laboratory.The method is processing the normal sperms with the swim-up and density gradient centrifuge process,and making six groups with finally the density being 0.1μmol/L,0.5μmol/L,1μmol/L,5μmol/L,20μmol/L,500μmol/L progesterone solution.The sperm and the unfertilizedⅡ,Ⅲoocytes are incubated together for 2 horns after incubating the six groups of sperm for 3.5 hours. To transfer the oocytes into new medium,and beat upon the sperms binding loosely onto the ZP gently by the inside diameter 250μm sucker.Move the oocytes to 4μl bubble under oil,and beat upon the sperms binding closely on the ZP,again calculate the total number of the sperm.Uses SPSS 11.5 to carry on the T-test and the variance analysis,carries on statistics processing to the result.The result is that 20μmol/L, 5μmol/L have the remarkable enhancement function in proportion to sperm binding oocytes compared with the blank group after the progesterone processing sperm 3.5 hours,and 500μmol/L instead drops,in 3.5 hours process is even the extremely remarkable drop,with density in 1μmol/L,0.5μmol/L,0.1μmol/L the progesterone group and the blank group compared with P>0.05,in statistics do not have the difference.The conclusion is the progesterone density when 20μmol/L,5μmol/L,may strengthen the sperm capacitation to a certain extent,and improve the ability of sperm binding to oocytes when 500μmol/L,the excessively high progesterone density obviously reduces the human's sperm acrosome membrane and the protoplasmic membrane fluidity,and causes the membrane member accumulation,instead strengthens the membrane stability,thus reduces the acrosome reaction,causes the sperm capacitation weaken,reduces the oocytes binding force.The density in 1μmol/L,0.5μmol/L,0.1μmol/L and so on low level's progesterone group and the blank group compares P>0.05,in statistics does not have the difference,explained that under the low concentration progesterone,the influence of progesterone to sperm capacitation and acrosome reaction be very small or nothing.These explains that progesterone only will proceed sperm capacitity an acrosome reaction in certain scope. Progesterone as one of main hormones in female animal gential tract,is very important for sperm capacitation and acrosome reaction,studies of these hormone representation-progesterone action mechanism,is advantageous to know other factor's mechanism,may provide theories for the development of Assisted Reproductive Technology(ART),and is also advantageous to raise fertilization rate and the quality of implantation embryos.
