【Objective】1 To evaluate the method to detect concentrations of SCu,SMn,SSe by microwave digestion- atomic absorption spectrometry(SCu detected by FAAS, SMn detected by GFAAS,SSe detected by HGAAS).2 To ascertain preliminarily the serum concentration reference values of copper, manganese and selenium of healthy adults in Jinan.3 To analyze the influences of sex and age to the values of SCu,SMn, SSe.4 Describe copper,manganese,selenium concentrations of soil,grains, vegetables,fruits,fresh meats,to study distributions of trace elements in environment.5 To investigate the influences of education,social-economic,tobacco smoking, alcohol consumption,physical exercise and sleep hours to serum copper, manganese,selenium concentrations.【Method】1 Determin two blanks everyday for six days,then calculate the standard deviation of these blanks and detection limits of the method.Determine high, midst,low standard solutions repeatedly to evaluate the precision of this method. Evaluate the method accuracy by detecting standard material and standard addition recovery.2 1005(423 male,582female) healthy physical examination subjects were recuited by cluster random sampling from every districts of Jinan.They were divided into 4 groups by age:652 responses for the interval of age 18~29,172 for 30~39,131 for 40~49 and 50 for the interval of 50~60yr,and the information of subject were obtained by questionnaire.3 Serum samples were digested by microwave,and wet digestion method was used to deal with soil,grains,vegetables,fruits and fresh mests.4 Copper was detected by flame atomic absorption spectrometry,mangaese was detected by graphite furnace atomic absorption spectrometry,selenium was detected by hydrogenation atomic absorption spectrometry.5 All analyses were performed by using the statistical analysis system SPSS11.5 and SAS9.1.One-way analysis of variance(ANOVA) was used to evaluate differences in the mean values of difference groups.Spearman correlation was used when analyzing the correlativity between age and serum trace elements concentrations.General linear model analysis(GLM) was applied as multivariate analysis.【Result】1 Detection limits of copper,manganese,selenium were 0.0023 mg/L, 0.0307μg/L,0.1411μg/L respectively.Precision was less than 7%and the average of standard addition recoveries was 92.54%.Detection value was unanimous to the true value of standard material.Serum reference values of Cu,Mn and Se(P2.5~P97.5) were 0.40~1.42 mg/L, 0.79~13.29μg/L,35.87~87.81μg/L respectively.3 The concentrations of serum Cu of female was higher than that of male (P<0.05),and raised by aging(r=0.14,P<0.01),especially for male(r=0.18, P<0.01).There were no differences of serum Mn between sex,although SMn raised by aging,but it was not statistically significant(P>0.05).The serum selenium level of male was higher than that of female(P<0.05),but the serum selenium level was not influenced by age.4 Average concention of vegatables were:Cu 0.88mg/kg,Mn 2.78 mg/kg,Se 0.06 mg/kg.Average concentration of fruits were:Cu 0.40 mg/kg,Mn 2.26 mg/kg, Se 0.11 mg/kg.Average concentration of grains were:Cu 4.83 mg/kg,Mn 11.16 mg/kg,Se 0.50 mg/kg.Average concentration of fresh meats were:Cu 0.70 mg/kg,Mn 0.72 mg/kg,Se 0.38 mg/kg.Average content of soil was:Cu 24.18 mg/kg,Mn 449.70 mg/kg,Se 0.87 mg/kg.5 Tobacco smoking and alcohol consumption seemed to degrade SCu concentration,especially for male.Smoking seemed to degrade SMn concentration,and alcohol consumption can degrade male SMn concentration. Alcohol consumption can degrade female SSe concentration,and smoking seemed to elevate SSe concentration.The relationships of education, social-economic,physical exercise,sleep hours and SCu,SMn,SSe content was not statistically significant.【Conclusion】Microwave digestion- atomic absorption spectrometry showed several advantages to detect serum trace elements,for example high standard addition recovery,precision and accuracy with satisfactory results.Serum copper and Se level in Jinan residents were lower according to other regions.Cu,Mn,Se contents of Jinan soil were moderate,but vegetables and grains content were lower than other regions.Tobacco smoking and alcohol consumption seemed to degrade SCu concentration,especially for male.Smoking seemed to degrade SMn concentration, and alcohol consumption can degrade male SMn concentration.Alcohol consumption can degrade female SSe concentration,and smoking seemed to elevate SSe concentration.Education,social-economic,physical exercise,sleep hours did not affect serum Cu,Mn,Se contents significantly.
