Study Of The Immunologic Protection On The Sarcoma-180 Bearing Mouse By Cartilage Polysaccharide

To investigate the immunological influence of Cartilage Polysaccharide that was extracted from cartilage of porcine by using sarcoma 180(S180) solid tumor and ascite tumor animal model in vivo and discuss its mechanism.S180 mice model was established by i.p. injection.The life span of S180 ascite tumor-bearing mice, index of thymus and index of spleen was increased by injecting cartilage polysaccharide i.p.. HE staining could detect changes of cells of spleen. We detect cellmediated immune response of tumor bearing mice. Lymphocyte transformation assay by MTT method showed that the stimulation indexes of T cells and B cells were both increased. The activity of NK cells of peripheral blood and spleen from tumor-bearing mice were detected at the 11th day after treatment. Erythrocyte rose forming cell test (ERFC) also used to detected immunologic function. The cartilage polysaccharide could significantly promote the activity of NK cells of tumor-bearing mice, enhance the rate of erythrocyte rose forming.Immunofluorescent assay demonstrated that cartilage polysaccharide could also increase the expression of CD40, CD40L in splenocytes of tumor-bearing mice.FCM data showed lymphocyte subsets changes : CD3~+, CD4~+ percentages of lymphocyte and rate of CD4~+/CD8~+ was increased after injecting cartilage polysaccharide. Phagocytic index of neutral red and index of chick erythrocyte phagocytosis of macrophage was also increased by injecting cartilage polysaccharide. Wright-Giemsa staining of blood cells show that determination of PMN of peripheral blood cells in mice was increased which could play important antitumor activity.Humoral immunity was detected by single immunodiffusion Test and ABC-EL/SA which was used to test cytokine of peripheral blood. Single immunodiffusion test show that IgG antibody significantly promote which could enhance humoral immunity response. Cartilage polysaccharide influence tumor-bearing mice secrete IL-2,IL-6, TNF-αon the mice serum level which regulate the tumor happen and develop and have positive effect on inhibiting the growth of S180 cells in vivo.In conclusion, cartilage polysaccharide can improve immunologic function of tumor-bearing mice. It is a novel antitumor agent.
