Leonurus,additional name, is herba leonuri or motherwort herb, belonging to the labiatae motherwort genus, its main active substances for alkaloids, flavonoids, terpenoids, fatty acid, naphtha, etc. Traditiontional Chinese medicine holds that leonurus can remove blood stasis, regulate menstrual and water, known as "the veins of rejuvenation""the effective prescription of multiparity ". Clinically, commonly used to treat abortion hemorrhage, coronary heart disease, myocardial ischemia, high blood viscosity, dysmenorrhea, kidney disease, and other skin diseases. Literature indicates that, motherwort has a definite effect on uterine bleeding after drug abortion , its effective ingredients are alkaloids such as leonurine, hydrochloric stachydrine, etc. Leonurine hydrochloride(Leo) is an new artificial synthetics and its structure is identical with natural leonurine. For its pharmacological effects, this subject investigated effects and mechanism of Leo on incomplete abortion rat uterus model induced by mifepristone(8.3mg/kg) and misoprostol (100mg/kg).The main contents were divided into some sections as follows:1. Effects of Leo on incomplete abortion induced by medicine in ratsSD early pregnancy rats were randomly divided into normal group, model group, Leo (0.5, 1.5, 4.5 mg/kg) group, Yimucaopian (22.5 mg/kg) group, Gongxuening capsule (130 mg/kg) group, each group of eight. At the seventh day of pregnancy, except for the normal group, every groups were induced incompletely abortion model by mifepristone compatibility misoprostol. The next day, collecting uterine bleeding, recording hemorrhage time and each group was drenched corresponding medicine and physiological saline respectively for 7 days successively. At the fifteenth day, rats were executed and its blood, hypothalamus, pituitary, and uterus were got with the conventional method for inspection. Results show that compared with the model group, Leo degraded the weight and index of uterus, decreased the amount of uterus haemorrhagia, shortened bleeding time, enhanced uterine contraction, elevated the level of serum estradiol(E2); Uterus histopathological examination showed Leo could also ameliorate tissue structure and reduced the residual of intra-uterus obviously by hematoxylin-eosin(HE) staining. These results suggested Leo has positively therapeutical effect on uterine bleeding induced by medical abortion probably by the enhancement of uterine contraction and the elevation of serum E2.2. The part mechanisms of therapeutic effects of Leo on incomplete abortion in ratsFurther study showed compared with the model group, Leo elevated rat serum level of follicle stimulating hormone (FSH) and luteinizing hormone(LH)after drug-abortion; Leo treatment up-regulated the expression of protein gonadotropin releasing hormone(GnRH) in hypothalamus and down-regulated protein laminin( LN) in uterus tissue by immunohistochemistry. By RT-PCR analysis, Leo treatment augmented the expression of estrogen receptor alpha(αER) and estrogen receptor betal (βER) mRNA in uterus tissue, but PRmRNA has no obvious change in uterus by Leo. In addition, endothelin(ET) and endothelin / nitric oxide (ET/NO) in uterus tissue were also raised by Leo. Conclusion: Leo has a significant therapeutical effect on abnormal uterine bleeding induced by medical abortion in rats,which result from the enhancement of uterus contraction, up-regulation of GnRH protein in hypothalamus andαER andβER mRNA in uterus tissue,depression of LN protein, elevation of serum E2 and ET/NO in uterus tissue, and regulation of hypothalamus-pituitary- ovarian axis function.
