Objective:To investigate the concentration of serum leptin and the disposition of Gln223Arg polymorphism in leptin receptor gene of patients with Gestational diabetes mellitus(GDM).The objective is to assertain the relationship between leptin, Gln223Arg polymorphism and GDM.Methods:1.Levels of serum leptin were determined by Radio immunoassay (RIA) in 62 patients with Gestational diabetes mellitus(GDM) and also in 60 normal pregnant women.2.Using polymerase chain reaction-restriction fragment length polymorphism method(PCR-RFLP) to examine the Gln223Arg polymorphism in leptin receptor gene among 62 patients with Gestational diabetes mellitus(GDM) and 60 normal pregnant women.Results:1.The serum leptin levels in pregnant women with GDM and normal pregnant women were 14.2±4.3μg/L and 10.1±2.6μg/L,respectively,suggesting the serum leptin levels were significantly higher in pregnant women with GDM than that in normal controls(P<0.05).2.The frequencies of GG,(GA+AA) genotypes of leptin receptor gene Gln223Arg were 61.29%,38.71%in GDM group,respectively,and they were 81.67%,18.33%in normal group,respectively(P<0.05).The frequencies of two alleles G and A of leptin receptor gene Gln223Arg were 77.42%,22.58%in GDM group,and 89.17%,10.83%in normal group.The frequencies of the allele A was higher in GDM group than that in normal group(P<0.05).3.The serum leptin level was15.3±4.1μg/L in GDM group with GA+AA genotypes.It was 13.5±3.7μg/L in GDM group with GG genotype(P<0.05).Conclusion:1.The levels of serum leptin were higher in pregnant women with GDM than that in normal controls.Serum leptin was correlated with GDM.2.There were G/A polymorphisms of leptin receptor gene Gln223Arg position nucleotide in Shandong province of china.The frequencies of(GA+AA) genotypes of leptin receptor gene Gln223Arg were higher in GDM group than that in normal group. Gln223Arg polymorphism in leptin receptor gene possiblely was association with GDM.3.In GDM group,The levels of serum leptin were higher in pregnant women with genotypes GA+AA than the pregnant women with genotype GG.The Gln223Arg polymorphism in leptin receptor gene could affect the level of leptin in GDM group.
