Changes Of Effective Components And Antidepressant Effects Of Radix Bupleuri And Radix Paeoniae Alba Before And After Vinegar Roasting

In Chinese medicine,Bupleuri Radix and Paeoniae Radix Alba have been shown to have significant therapeutic effects on "depressive symptoms".The compatibility of Bupleuri Radix and Paeoniae Radix Alba can soothe the liver and relieve depression,so that the liver qi can be regulated.It is a king’s medicine;The medicine for soothing the liver and regulating qi must cooperate with the official medicine for nourishing blood and blood-white peony to fundamentally solve the problem of liver depression.Therefore,this study was conducted to explore the changes in chemical composition of Bupleuri Radix before and after vinegar roasting with Paeoniae Radix Alba,and the effects of Bupleuri Radix before and after vinegar roasting with Paeoniae Radix Alba on the antidepressant effects in mice.The following studies were made for this experiment:1.Study on the changes of compatible components of Bupleuri Radix and Paeoniae Radix Alba before and after vinegar roasting Firstly,Bupleuri Radix was roasted with vinegar according to the method in the Chinese Pharmacopoeia.According to the clinical medication ratio,bupleurum was compatible with white peony in three proportions(1:2,1:1 and 2:1)before and after vinegar roasting.The content differences of main components in different proportions were compared to study the changes of compatible components between Bupleuri Radix and Paeoniae Radix Alba before and after vinegar roasting.The results showed that the content of saikosaponin a decreased and the content of saikosaponin b2,paeoniflorin and paeoniflorin increased when Bupleurum and Paeonia Alba were mixed in the ratio of 1:2,1:1 and 2:1.2.Comparison of the fingerprints of Bupleuri Radix and Paeoniae Radix Alba(1:2)with those of Radix Bupleuri and Radix Paeoniae Alba(1:2)in vinegar The fingerprint profiles of(vinegar)Bupleuri Radix and Paeoniae Radix Alba were established,and cluster analysis,principal component analysis and partial least squares-discriminant analysis were performed on the fingerprint profiles of Bupleuri Radix and Paeoniae Radix Alba before and after vinegar roasting,it showed that there were significant differences in the fractions before and after vinegar roasting,and four potential quality markers were identified,indicating that the vinegar roasting of Bupleuri Radix had an effect on the changes in the content of the components of the Bupleuri Radix and Paeoniae Radix Alba pairing.3.Study on the antidepressant effect of Bupleuri Radix before and after vinegar roasting with Paeoniae Radix Alba(1:2)and the tablet dosage form After different ratios of different doses of(vinegar)Bupleuri Radix-Paeoniae Radix Alba decoction were studied in mice’s voluntary activity experiment,tail suspension experiment and forced swimming experiment,the acute depression model was created with Risperdalin,and the content of neural factors in the brain of mice with different extracts was studied.In the determination of behavioral and brain neural factors,the combination of Bupleuri Radix vinegar roasted with Paeoniae Radix Alba could shorten the time of voluntary activity and immobility,tail suspension and immobility,swimming and immobility,increase the content of 5-hydroxytryptamine and dopamine in the brain,and restore the brain norepinephrine to normal level.Studies have shown that the antidepressant effect of Bupleuri Radix vinegar roasted with Paeoniae Radix Alba(1:2)is more pronounced.The vinegar decoction of Bupleuri Radix-Paeoniae Radix Alba(1:2)was selected to screen the ratio of different fillers,add disintegrants as well as binders,etc.for tablet process study.The quality of three batches of vinegar Bupleuri Radix-Paeoniae Radix Alba tablets was examined,and the results showed that their properties,weight difference,hardness,fragility and disintegration were all qualified.
