Objective:Acute necrotizing pancreatitis (ANP) has seriously endangered the life and health of human beings, and it is often accompanied with multiple organ dysfunction syndrome(MODS). ANP complicates with hepatic injury as high as 88.9%, which would not only increase the ANP disease, but also affect the prognosis. With the deep study on the pathogenesis of ANP mechanism, it is demonstrated that macrophages play an important role in antigen presentation-activated immune responses by releasing large quantities of chemical and inflammatory cytokines of the liver and multiple organ injury of ANP patients. This paper is to investigate the protective of lipsomal clodronate on hepatic injury in rats with acute necrotizing pancreatitis (ANP) by importing lipsomal clodronate into macrophages in ANP rats.Method:Firstly, Lipsomal clodronate is prepared by means of thin film. The models of ANP were established by injection of sodium taurocholate into the pancreatic capsule. The ANP models were prepared and randomly assigned to the model control group(C), ANP+ Liposome group(P), ANP+ Liposomal Clodronate group(T). The serum levels of IL-6, TNF-αwere measured, and the changes of ALT, AST and AMS in serum were determined. Pathologic alteration of pancrease and liver were observed and the pathological score of pancreatic tissue was also estimated.Result:In P group, serum levels of ALT,AST,AMS were significantly elevated compared with C group (P<0.01). In T group, serum levels of ALT,AST,AMS were significantly decreased as compared with P group (P<0.01). In P group, serum levels of IL-6,TNF-αwere significantly elevated as compared with C group (P<0.01). In T group, serum levels of IL-6,TNF-αwere significantly decreased as compared with P group (P<0.01). In T group, pathologic damages of liver and pancreas were significantly decreased as compared with P group and pancreatic tissue pathological score were lower than those in P group(P<0.01).Conclusion:It was feasible to prepare lipsomal clodronate by means of thin film.Excessive release of inflammatory mediator play an important role in liver injury of ANP rats. We found that liposomal clodronate could depress the function of macrophages in ANP rats and it could alleviate the liver injury of ANP rats.
