Depression is a kind of mental disorders which often be overlooked. San Sheng He Huan Yin granule, a clinical prescription which including 12-herb Chinese herbal is proved to have the exact effect on the treatment of depression after a long period of clinical use. Because of the big formular, it is particularly important to research its active ingredients and to establish scientific and reasonable quality control standards. Frutus aurantii and rhizoma anemarrhenae are belongs to the gentleman medicine, there have been reports about the efficacy of the the treatment of depression. HPLC was employed to establish a double passage simultaneous determination metheod. Several active ingredients was determined qualitatively and quantitively.The first part summarizes the research of depression, material research, and overviewed the serum pharmacochemistry of Chinese medicine. A detail description is made to introduce the composition of San Sheng He Huan Yin granule and the current research on pharmacological research and analysis methods.In the second part, San Sheng He Huan Yin granule was analysised by HPLC coupled with the same reference standard retention time and UV absorption spectra of particles, which proved the San Sheng He Huan Yin granule obtained mangiferin, sinapine, naringin, hesperidin, neohesperidin and timosaponin B-Ⅱ, etc. Meantime the ownership of single herb was examined and HPLC conditions were groped. The active ingredient above is studied by serum pharmacochemistry method. It is proved that Sinapine, naringin, hesperidin and neohesperidin can be absorbed into blood. According to the literature, mangifern and timosaponin B-Ⅱ also can be absorbed into the blood, and take clinical effect. So that the above 6 compounds were choosed as active ingredients.The third part of the paper established a two simultaneous determination method of DAD detector and ELSD detector for examining the composition of the 6 active ingredients, the method can simultaneously detect the 6 components, which is simple, rapid, accurate and reproducible.
