Tongsaimai pellets is a new compound preparation,secondary development based on the the famous Chinese medicine Tongsaimai Tablet.Tongsaimai pellets is composed of six herbals including Flos Lonicera japonica, Radix Angelica sinensis, Radix Astragali,Radix Scrophulariae, Radix et Rhizoma Glycyrrhizae.which with the complexchemical composition, including volatile oil,flavonoids, saponins, phenolic acids and other active ingredients of several major categories.Preliminary studies have been showed the effective composition of the new compound preparationfor the treatment of ischemic stroke,and new durg have been made in accordance with the requires of category 5 of chinese medicine.The main task of this topics is Establishment of chromatographic fingerprint and serum pharmacochem istry of TongSaiMai pellets.Focus on the vitro and in vivo changes of chemical composition of Tongsaimai Pellets, lay the foundation for the further study the material basis of Tongsaimai pellet to the treatment of ischemic stroke.The study comprises three parts:The first part is Establishment of chromatographic fingerprint of Tongsaimai pellets, divided into three sections including HPLC-UV fingerprint, UPLC-UV fingerprint and their comparison.The second part is the chemical analysis about Tongsaimai pellets in vivo and vitro.Including in vitro Tongsaimai pellets chemical analysis, determination of index components and composition of serum chemistry analysis and comparison of in vivo differences between the chemical composition and possible causes. The preliminary analysis of 46 chemical compositions of Tongsaimai pellets have been made by UPLC/MS-Q-TOF technology.And next is the study of serum pharmacochemistry of Tongsaimai pellets. According to research methods of serum pharmacochemistry,as rats for experimental animal, and prepare serum contained bushentiaochong after the rats are dosed by irrigating stomach, applying the method of HPLC-UV compare the Tongsaimai pellets raw liquor, The serum contained medicine after the rats and blank serum,questing chromatographic peak of migration from blood, and according to the relative retention time and online UV spectrogram of each peak, deduce the medicinal materials where the the migration compositions in blood come from and the attributive compounds, by contrast to the serum contained medicine of each simple preparation.And further structural analysis of tha the blood components have been made by UPLC/MS-Q-TOF technology and 23 components into the blood be identified.The third part is the text of the conclusion, with a view to the research of fingerprint pharmacodynam ics of traditionalChinesemedicines and analysis of the chemical composition in vivo-in vitro correlation, bring forward the next phase of the proposed research prospects.
