Chronic fatigue syndrome(CFS)is a kind of syndrome that profoundly manifested in long term of extreme exhaustion, which occurs mostly among women at the age of 20 to 50. It belongs to the category of sub-health, which concerns mostly about the human health in the 21 century.It belongs to deficiency and exhaustion, insomnia, palpitation,lily disease in traditional Chinese medicine(TCM). As the pathogenic factor and etiopathogenesis of the disease is unclear up till now, there are not effective measures for prevention and treatment. Research of modem medicine on CFS mainly manifests in its etiopathogenesis and the method of model preparation.It is commonly accepted that CFS is related with long-term spiritual stimulation, excessive exhaustion of brain and physical force, and virus infection, accompanying with malfunction of nerve-endocrine-immune system. There are two methods in model preparation:one is" chronic bondage".another is the complex of chronic bondage and swimming in cold water, accompanying with electrical stimulation while eating. According to the above two methods and learn from other's experience, this experiment adopted the method of chronic bondage and swimming in the cold water alternately in model preparation, which proved to be reasonable and convenient.Acupuncture and moxibustion is a treatment that regulates the function of the neural and endocrine system of the body and reinforces the immune function and the capability of preventing diseases. It is prior in wholly regulation, and has the advantage of easy manipulation and convenience, no side effect, being economical, effective and safe. So, on the basis of a great quantity research of antiquity and modem literature, this text demonstrated the theoritical evidence of acupuncture to therapy CFS, and designed and put this experiment into practice. 1. PurposeWe try to reveal the regulated mechanism of acupuncture through observing the change of weigh and behavior and the index such as TGF-βand IL-1β,according to random and antithetical principle.2. MethodRandomly divided the animals into model group, acupuncture group, ginsenoside group and normal control group. We got the rat model of chronic fatigue by the method of chronic bondage and swimming in cold water alternately. The acupuncture group used the acupoints of Baihui, Shenshu, zhusanli, according to Experimental Acupuncture Science, alternately on both sides. Ginsenoside group was stomach fulfilled by ginsenoside. The normal control group was not treated. Four weeks later, measured the behavior of rats, recorded the weigh of the rats, then had their head cutted and blood sampled. Recorded the weigh of the hypothalamus and pituitary, then measured TGF-β1 and IL-1β.3. Results(1)Weigh:As time goes, the model group ate less and drank less. Their hair lost luster and they lost weigh obviously. There is remarkable difference compared with the normal control group. (P<0.01);while the acupuncture group gained weigh more than the model group, and there are statistics difference(P<0.05).(2)Change of behavior:The measure of open-field activity showed that, the model group had distinctive decrease in horizontal and vertical motion when compared with the normal control group(P<0.01). The experiment of tail-hanging showed that, the still period after tail-hanging of the model group was obviously longer than the other groups;The still period of the acupuncture group was decreased, and the difference was distinctive compared with the model group.(3) TGF-β1 and IL-1βin serum:the model group was obviously higher than the normal control group, ginsenoside group and the acupuncture group (P<0.01). The acupuncture group had no statistics difference compared with the normal control group, but different from the model group (P>0.05 and P<0.01 respectively).4. Discussions(1) Acupuncture increased the weigh of model rats, elevated their emotional responses and seeking activities, which proved that, acupuncture improved the physical force of chronic fatigue rats and excitability of their neural system, increased their spontaneous activity.(2) Acupuncture decreased the content of TGF-β1 and IL-1βin serum of model rats, suggesting its function in resisting the capability of responding to stimuli and fatigue may be related with its adjustment to the immune cells, easing the damage of the chronic fatigue rats'immune function, or may be by beta cells factor TGF-β1 and IL-1βbeta as a media to regulate the function of the nerve-endocrine system to implement the mechanisms, which need to further study.
