The normal microbial flora of the vagina plays an important role in preventing genital and urinary tract infections in women. Thus an accurate understanding of the composition and ecology of the ecosystem is important to understanding the aetiology of these diseases. At present common wisdom about microbial flora of the vagina is based on methods of microscopic examination and traditional cultivation of microbial populations; however, it leaves many bacteria uncultured and unidentified, which are closely linked with genital diseases.In this study vaginal swabs collected from different diseases(babterial vaginosis, cervical cancer, infection of HPV, Cervical intraepithelial neoplasia degree 1 and 2-3, cervical erosion), Total microbial community DNA was isolated following resuspension of microbial cells from vaginal swabs, cultivation-independent methods PCR-DGGE were used to analysis the composition of vaginal microbial community, in DGGE gel special bands were cutted and sequenced,composition of microflora community were identified. The main results were as follows:1. The findings indicate the diversity and kinds of organisms that comprise the vaginal microbial community varied among women, further diversity in the etiological agents associated with BV. At the phylum level, Actinobacteria and Bacteroidetes were strongly associated with BV.The dominance by Lactobacillus,the presence of several novel species(Prevotella, Gardnerella, Atopobium).especially higher levels\of A.vaginae in disturbed vaginal microflora-indicate that much is to be learned about the composition of the vaginal microflora and its relation to the etiology of BV. Prevotella may be associated with a variety of diseases.2. DGGE profiles showed that bacterial species diversity in bacterial vaginosis and cervical erosion2°specimens is high, bacteria DNA structure is complex.Mycoticvaginitis and colpitis specimens's bacterial species diversity is secondary to bacterial vaginosis, which consistent with the performance of vaginal infections.3. Tthere are similar community structure between sample of cervical erosion in degree 1,2 and 3; cervical cancer and CIN2-3 samples have another similar community structure.Thus, this study illuminates the complex structure of the vaginal ecosystem and calls for the consideration of microenvironments when sampling vaginal microbiota as a clinical predictor of vaginal health. The results showed will help researchers to grasp the various pathological conditions of microbial community structure, and identify related pathogens by designed species-specific primers, also provide a scientific basis for clinical diagnosis and treatment.
