Objective: By means of detecting HE4 and CA125 in serum of healthy women, benign ovarian tumor and ovarian carcinoma patients.To explore the possibility value of HE4 as tumor markers, compared withCA125, in the diagnosis of ovarian cancer.Methods: 1. The double antibody sandwich Enzyme-linked Immunosorbent Assay (ELISA) technique was applied to detect serum HE4 protein concentration, among this series,50cases healthy women,62cases benign ovarian tumor and ovarian cancer each 52 cases.CA125 concentration from Shanxi Province Cancer Hospital was detected by an electrogenerated chemilumin cence method. The levels of HE4 in each group were compared respectively. The correlations betweenserum concentration and different clinicopathological character in epithelial ovarian cancer tissue were analyzed.2.The sensitivity of HE4,CA125 in serum whether alone or combined in diagnosing ovarian carcinoma was compared. The diagnosis efficiency of the HE4 assay was compared, when sets different diagnosis points.3. Reference to benign ovarian tumor and Optimal cutoff values of serum HE4 as the standard point, Analysis the Value of HE4 alone and combined assay with CA125.4.All statistical analyses were performed with SPSS 13.0 statistical software. Results: 1.In healthy women, benign ovarian tumor, ovarian carcinoma, the serum HE4 median levels were 41.10,43.98and 260.90 (pmol/L) and CA125 median levels were 17.98,13.89 and 243.1 (U /ml).The median preoperative HE4 value of ovarian carcinoma group was significantly higher than each of the former two groups respectively(P<0.05). The median preoperative CA125 value of ovarian carcinoma group was significantly higher than each of the former two groups respectively(P<0.05) and the former two groups compared, was also statistically significant difference (P<0.05).2 In ovarian carcinoma group, the HE4 level in serum positively was associated with the concentration of CA125, but age was not. the HE4 level in serum of ascites group was significantly higher than without one(P<0.05),but FIGO stage, grade and lymphatic metastasis were no statistically significant difference respectively.3 Divided the HE4 level in serum into 50,75,100 and 150 pmol / L 4 level and ceiling, HE4 set point of 75 pmol/L. When exceed this point, diagnostic efficiency of ovarian carcinoma is improved. 4 The diagnosis efficiency of HE4 alone is better thanCA125 alone.Combined assay is better than CA125 alone.Conclusions:1 The serum HE4 level cou1d be seen as markers for screening of the ovarian carcinoma. CA125 and HE4 are complementary in screening the ovarian carcinoma, so they can improve the diagnosis efficiency.2 The HE4 level in serum of ascites group was significantly higher than without one(P<0.05),but FIGO stage, grade and lymphatic metastasis were no statistically significant difference respectively.3 The set point of 75pmol/L is advantageous for the improving the diagnostic sensitivity of ovarian carcinoma4 The diagnosis efficiency of HE4 alone is better thanCA125 alone. Combined assay is better than CA125 alone.
