The studied subject is Fufanghongdoushan granule, which is based on clinically proved recipe and composed under the theory of Chinese medicine.Fufanghongdoushan granule which is made from five kinds of herbs, such as Taxus chinensis, Radix Astragali, Herba Dendrobii, American Ginsen and Radix Glycyrrhizae. The granule is an effective prescription in treating cancer and improving immunity. For patients' convenient taking and clinical easy-using, we made a study on innovation of its formula, in order to do industrial large-scale production.In the studies on the preparation Proeess, orthogonal trial was used to select the optimal collection methed of collecting active ingredients from herbs.The water-insoluble composition of Taxus chinensis was extracted by percolation of ethanol. First,we selected the indexes to 10-de-acyl-baccatinⅢextraction yield as well as the chloroform extract as an inspection target. A lesser extent, we studied percolation rate, ethanol concentration Ethanol consumption, which were closely related to the yield of active ingredients, with L9(34) orthogonal. The results had showed that,9 times of 60% ethanol percolation, percolation flow rate as 7mL·min-1The dregs of Taxus chinensis after percolation, which have water-soluble composition of effective treatment of cancer, should be extracted by water. So, we put the dregs, Radix Astragali, Herba Dendrobii and Radix Glycyrrhizae together into the hot water for extracting water-soluble components. We took the yield of glycyrrhizic acid mono-ammonium salt as an indicator. With L9(34) orthogonal, the best extraction methed was like that,9 times of hot water extraction,2 times, each time 2 hours.After mixed the water extract and ethanol extract, we used spray-drying. According to the investigated the factors as inlet temperature, spray rate, and the liquid density, which were on the drying effects of water content of the finished product. With L9(34) orthogonal experimental table arrangements for the final optimal conditions:the density of extract was 1.15 g·mL-1, the feed rate of 30 mL·min-1, inlet temperature controlled at 180℃, as to achieve granulation requirements. In accordance with the proportion of American ginseng will be added to the preparation of prescriptions Then, we studied the dosage and category selection of the accessories. Comparison of soluble starch, carboxymethyl starch sodium, dextrin of dressing on the impact of hygroscopic particles, the results showed that, dextrin best to improve the particle hygroscopicity. At the same time, we determined the dextrin mixed with the dry extract mass ratio was 4:5. We also inspected the dosage of the correction deodorant—Stevioside, as well as the concentration of the wetting agent—Ethanol.The results showed that, dosage of stevioside as 2% of the total preparation was tasted better; with 85% ethanol concentration of the best.
