Objective: Comparison of amlodipine perindopril and hydrochlorothiazide combined with losartan on 2 essential hypertension the antihypertensive efficacy and safety.Method: 80 new cases of newly diagnosed or stop taking blood pressure medicine more than 2 weeks in patients with essential hypertension were randomly divided into two groups, 40 patients in each group. A (Amlodipine Perindopril) orally 1 time every morning, 4mg perindopril (goods Mingyashida, Servier Pharmaceuticals produced) and 5mg amlodipine (brand name Norvasc, Pfizer pharmaceutical company). B (Hydrochlorothiazide Losartan) orally 1 time every morningè12.5mg hydrochlorothiazide(Brand name double g, Qinhuangdao, North China Pharmaceutical Co., Ltd. Production)and 50mg losartan (brand name Cozaar, the U.S. pharmaceutical company Merck Sharp & Dohme). Treatment for 8 weeks, 8 weeks observation period is not combined with other antihypertensive drugs. Therapy in the treatment of 8 weeks before and 24h ambulatory blood pressure monitoring, respectively (ABPM) day average systolic blood pressure (24hSBP) and diastolic blood pressure (24hDBP), daytime average systolic blood pressure (dSBP) and diastolic blood pressure (dDBP), night mean systolic blood pressure ( nSBP) and diastolic blood pressure (nDBP), observed the impact of drugs on heart rate, adverse drug reaction, and serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), blood urea nitrogen (BUN), creatinine (Cr ), glucose (FPG), cholesterol (TC), triglyceride (TG),uric acid(UA), K~﹢.Result: (1) Amlodipine Perindopril group (A group) were compared with those before treatment after treatment, 24-hour mean blood pressure, daytime mean blood pressure, nighttime blood pressure decreased an average of 32.02/15.35 mmHg, 35.11/21.85 mmHg, 25.58/14.79 mmHg; Hydrochlorothiazide Losartan group (B group) were compared with those before treatment after treatment, 24-hour mean blood pressure, daytime mean blood pressure, nighttime blood pressure decreased an average of 32.11/14.99 mmHg, 35.82/20.81 mmHg, 26.00/14.41mmHg. The statistical analysis of the two groups were significantly decreased blood pressure after treatment (P <0.01), the antihypertensive effect of the two groups no significant difference (P> 0.05). (2) A, B groups before and after treatment heart rate, blood biochemical and other tests, no significant difference (P> 0.05). (3) A course of treatment group, ankle edema in 1 case, facial flushing in 1 case, a dry cough two cases; B group, 2 cases of dizziness, loss of appetite in 1 case, but tolerable without stopping. Side effects were 10% and 7.5%, no significant difference betweenthetwo(P>0.05).The conclusion : (1) of amlodipine + perindopril and losartan + hydrochlorothiazideantihypertensive effect exactly, no difference in antihypertensive effect between the two. (2) ofamlodipine + perindopril and losartan + hydrochlorothiazide on the heart rate, bloodbiochemistry had no effect on the indicators, no significant adverse reactions, and is effective,safe, reliable and convenient drugs.
