| OBJECTIVE (1)To analyse the characteristics of Han RhD negative blood donors with DEL phenotype in Fuzhou.(2)To research The molecular mechanism of DEL phenotypes in Fuzhou.METHODS DEL phenotypes were screened with absorption-elution test from 165 Han RhD negative blood donors in Fuzhou and tested serological RhCE phenotypes with anti-C,anti-c,anti-E,anti-e monoclonal IgM antibodies; It was adopted that the polymerase chain reaction with sequence-specific primer(sPCR-SSP)based on eleven pairs specific primers to amplify RHD exon 1~10 and hybrid Rh box. the exon 9 was analyzed by DNA sequences; the transcripts of DEL were analyzed by Reverse transcription-polymerase chain reaction(RT-PCR)and cDNA sequences. Chi-square test was used to analyze the different distribution of RhD negative blood donors with DEL among Changchun,Yunnan,Zhejiang,Shanghai,Taiwan and Fuzhou.RESULTS1. 48 DEL(about 29.09%)were found in 165 Han RhD negative blood donors in Fuzhou with absorption-elution test.while 117 Truly RhD negative about 70.91%2. Among 48 DEL phenotype blood donors, 36 for Ccee (75.00%), 10 for CCee (20.83%), 1 for CCEe (2.08%), 1 for ccee (2.08%); Among 117 Truly RhD negative blood donors, 85 for ccee (72.65%), 21 for Ccee 17.95%, 4 for CcEe (3.42%), 3 for ccEe(2.56%), 2 for CCee (1.71% ), 1 for CcEE (0.85%), 1 for ccEE(0.85%).3. All of 48 DEL phenotype blood donors carry complete 10 exon of RHD genes and RHD1227A allele, 43 hybrid Rh box were positive, turned out to be RHD1227A /RHd(89.58%), 5 hybrid Rh box were negative(11.63%),4 for RHD1227A/A, 1 for RHD1227G/A.4. All of 30 DEL blood donors have 2~4 different transcripts, Those are the transcripts with the deletion of exons7to 9(del789), exons7 and 9 (del79), exons 8 and 9(del89) and exon 9(del9). One of DEL have a transcript with (RHD1-2CE3-9D10) sequence, all of DEL blood donors did not have the normal transcripts..CONCLUSION DEL was accounted for about 29.01% in Han RhD negative blood donors in Fuzhou, the characteristic of RhCE phenotype of DEL was for Ccee (75.00%), Ccee (20.83%). All of DEL blood donors have RHD1227A Allele. RHD1227A may be an important genetic marker for DEL blood donors in Fuzhou. The molecular mechanism of the Han DEL blood donors in Fuzhou allele may be relate to RHD1227A Allele which cause all of DEL transcriptions with the deletion of exon9 and cannot express normal RhD protein. |
