Background:In the world wide, cancer is the third fatal killer after cardiovascular and infectious diseases. Gastric cancer is the second most common cause of death from cancer in Asia, And the most common cancer in China. The etiology of gastric cancer consists of genetic susceptibility, Psychological and environmental risk factors. The pathogenesis of gastric cancer is not yet fully understood. Recent studies showed that ADIPOQ polymorphism is significantly correlated with the risk of various cancers, meanwhile, the lastest research focused on gastric cancer reported that the plasma adiponectin level is correlated with tumor. But the association between ADIPOQ gene polymorphisms and gastric cancer risk is unknown. So we conducted a case-control study of gastric cancer among Chinese people, to explore the association of genetic variants of the adiponectin genes with gastric cancer risk. To understand the relationship between the incidence of gastric cancer and environment, we take analysis the geological and meteorological environment in Gansu province.Methods:We genotyped 199 gastric cancer cases and 232 controls for 2 haplotype-tagging single nucleotide polymorphisms(SNP) of ADIPOQ. Genotyping for 2 SNPs was performed by PCR-RFLP. In china, Gansu province is one of the highest incidence of gastric cancer area. To understand the relationship between the incidence of gastric cancer and geological and meteorological environment, we take analysis with the data of environment factors of 703 gastric cancer patients in Gansu province hospital from 1990/1/1 to 2007/12/31.Results:Heteroplasmy TG for rs2241766 was associated with decreased gastric cancer risk(p=0.038; OR,0.653; 95%CI,0.436-0.977). But in the analysis statified by gender,age,tumor differentiation and climatic region, this difference is abolished. The rs266729 SNP did not shown any significant differences. The gastric cancer patient mainly at 1680 to 1940 meter above sea level region with 6 to 8 average temperature, Temperate semiarid areas, sierozem and yellow loess soil as the main soil types.Conclusions:These findings suggested genetic polymorphisms of ADIPOQ SNP(rs2447166) were associated with significant alternative risk of GT in Chinese population. This is the first report of an association between variants of the ADIPOQ and gastric risk. The results warrant further studies of the adiponectin pathway in gastric cancer. The result also convinced that environment specificity of this region is one reason of the high incidence of gastric cancer in China.
